摘  要:目的:探讨间充质干细胞源性外泌体的成功提取方法。方法:应用试剂盒二次提取huc-MSCs外泌体,凝胶电泳法及BCA蛋白定量法检测两次提取外泌体蛋白纯度与含量的不同;电镜及流式细胞仪鉴定外泌体。结果:凝胶电泳法及BCA蛋白定量法证实二次提取后外泌体蛋白纯度提高,形态特征明显,并高表达CD63及CD81蛋白。结论:间充质干细胞外泌体经过二次提取纯化后,纯度提高,可以更准确的开展后续实验。Objective:To study the successful extraction method of mesenchymal stem cell derived exocrine.Methods:The exocrine of huc-MSCs was secondarily extracted by extraction kit,the purity and content of exocrine proteins were detected by gel electrophoresis and BCA protein quantification,and the exocrine was identified by electron microscope and flow cytometry.Results:Gel electrophoresis and quantitative analysis of BCA protein confirmed that the purity of exocrine protein was increased after secondary extraction.The morphological features were obvious and CD63 and CD81 proteins were highly expressed.Conclusion:The exocrine of mesenchymal stem cells after secondary extraction and purification can improve the purity of the exocrine of mesenchymal stem cells,and the follow-up experiment can be carried out more accurately.
