摘 要:目的:检测碳量子点对神经细胞的生物成像效果,及其生物安全性评估。方法:使用动态光散射法和荧光分光光度计对碳量子点进行表征,使用共聚焦显微镜观察碳量子点的生物成像效果,用MTT法检测碳量子点对细胞的毒性效应。结果:碳量子点光学效应好,激发光为375nm时,碳量子点荧光强度最强,而且碳量子点荧光强度与其浓度正相关。碳量子点可被PC12细胞摄取并发出明亮的荧光,显示出良好的荧光成像效应。同时发现,碳量子点对PC12细胞毒性低,500μg/m L碳量子点孵育48h后,细胞活力仍保持在70%以上。结论:碳量子点荧光成像特性优异,生物安全性好,在口腔医学领域应用时不会造成神经系统损伤。Objective: To evaluate the biological imaging effect and the biocompatibility of carbon quantum dots(CDs)on neurons. Methods: The hydrodynamic size and optical performance of CDs in distilled water(DW) were respectively determined by dynamic light scattering(DLS) and fluorescence spectrophotometer. Furthermore, cell survival rates following treatment with CDs were evaluated using the MTT assay and the biological imaging effect of CDs was observed under a confocal microscopy. Results: CDs shows excellent optical effect, the peaking fluorescence intensity of CDs was observed when the wavelengths at 375 nm, and the fluorescence intensity of CDs was dose-dependent. CDs could be taken up by PC12 cells and emitted bright fluorescence, which showed a good fluorescence imaging effects. At the same time, it was found that CDs had low toxicity to PC12 cells. After PC12 cells were incubated 48 h with 500μg/m L CDs, cell viability remained at 70% or more. Conclusion: CDs have excellent fluorescence imaging properties, good bio-safety, and do not cause neurological damage when applied in the field of stomatology.
