摘 要:目的用髓鞘少突胶质细胞糖蛋白35-55(MOG)多肽免疫C57BL/6小鼠建立实验性自身免疫35-55性脑脊髓炎(EAE)模型,探讨EAE模型发病期与相关免疫指标的关系。方法用MOG抗原免疫C57BL/6雌35-553性小鼠,观察其临床症状和中枢神经系统的病理改变;应用H方法测定EAE小鼠脾脏T淋巴细胞活化增殖程度;++应用流式细胞术检测中枢及外周脾脏淋巴细胞中CD4 CD25调节性T细胞;荧光定量RT-PCR检测IL-17A、Foxp3、IFNγ的表达水平。结果成功建立EAE模型。与对照组相比,EAE组中可见小鼠中枢神经系统有淋巴细+++胞浸润,脾脏T淋巴细胞增殖随MOG多肽浓度增加而增强,CD4 CD25调节性T细胞占CD4 T细胞的比例明显降35-55低(P<0.01),IL-17和IFNγm RNA表达量显著升高(P<0.05),Foxp3 m RNA表达量显著降低(P<0.05)。结论 MOG多肽成功诱导了C57BL/6小鼠EAE模型,Th17与Treg细胞共同参与调控EAE的病理过程。immunization with myelin oligodendrocyte glycoprotein 35-55(MOG) peptide in C57 BL/6 mice and explore the 35-55 relationship between the incidence of EAE and related immune indicators.Methods Female C57 BL/6 mice were immunized with MOG antigen,and the clinical symptoms and pathological changes of central nervous system were 35-553 observed,H method was used to determine the degree of activation and proliferation of spleen T lymphocyte in EAE mice;+ +CD4 CD25 regulatory T cells were detected in the central and peripheral lymphocytes by flow cytometry;expressions of IL-17 A,Foxp3 and IFNγ were detected by fluorescent quantitative RT-PCR.Results EAE model was established with lymphocyte infiltration in the central nervous system of EAE group.The proliferation ability of T lymphocytes increased with + +the increase of MOG polypeptide concentration in spleen of EAE group.The percentage of CD4 CD25 regulatory T cells 35-55 was significantly lower,the expression levels of IL-17 and IFN gamma m RNAs were significantly higher,and the expression level of Foxp3 m RNA decreased significantly in EAE group than in control group(P〈0.05).Conclusion C57 BL/6 EAE model was successfully constructed by MOG peptide induction,Th17 and Treg cells might be involved in the 35-55 pathogenesis of EAE.Objective To establish an experimental autoimmune encephalomyelitis(EAE) model by
