摘 要:目的探讨转录信号转导子与激活子-3(STAT3)和Hedgehog干细胞调控信号通路相关蛋白对乳腺癌多柔比星耐药性的影响。方法自2016年1月至2017年1月,前瞻性收集40例乳腺癌组织标本和乳腺癌旁组织标本,比较乳腺癌组织和癌旁组织中STAT3、乙醛脱氢酶(ALDH1)、Smo蛋白和Gli1蛋白表达的差异,同时比较人多柔比星敏感和耐药乳腺癌细胞系:MCF-7和MCF-7/ADR中STAT3、ALDH1、Smo蛋白和Gli1蛋白表达的差异。结果在乳腺癌组织中STAT3(-)、(+)、(++)和(+++)分别占7. 50%、17. 50%、50. 00%和25. 00%,在癌旁组织中分别占30. 00%、45. 00%、17. 50%和7. 50%,差异有高度统计学意义(P <0. 01)。多柔比星耐药乳腺癌细胞中STAT3(-)、(+)、(++)和(+++)分别占5. 00%、15. 00%、50. 00%和30. 00%,多柔比星敏感乳腺癌细胞中分别占30. 00%、45. 00%、15. 00%和10. 00%,差异有高度统计学意义(P <0. 01)。乳腺癌组织中ALDH1、Smo蛋白和Gli1蛋白表达均高于癌旁组织(P <0. 05),且在多柔比星耐药乳腺癌细胞中表达均高于多柔比星敏感乳腺癌细胞(P <0. 05)。结论 STAT3和Hedgehog干细胞调控信号通路相关蛋白在乳腺癌中高表达,可能与多柔比星耐药有关。AIM To explore the correlation between signal transducers and activators of transcription( STAT3 ) and regulatory signaling pathway related protein of stem cell Hedgehog on doxorubicin resistance of breastcancer. METHODS Totally 40 tissue specimens of breast cancer and tissue specimens beside breast cancer fromJanuary 2016 to January 2017 were prospectively collected, and STAT3, acetaldehyde dehydrogenase 1 ( ALDH1 ) , difference between Smo protein and Glil protein in the tissue specimens of breast cancer and the issue specimens beside breast cancer were analyzed, while doxorubicin sensitive breast cancer cell line with and doxorubicin resist-ant breast cancer cell line were compared : STAT3, ALDH1, and the difference between Smo protein and Glil pro-teininMCF-7 andMCF-7/ADR. RESULTS STAT3 (-), (+), ( + +) and ( + + +) in breast cancertis-sues were respectively 7.50% , 17.50% , 50.00% and 25.00% , while the ones in tissues beside breast cancerwere respectively 30.00% , 45.00% , 17.50% and 7.50% , and the difference was statistically significant ( P 〈0.01 ). STAT3 ( - ), ( + ) , ( + + ) and ( + + + ) of doxorubicin resistance breast cancer cells were 5.00% ,15.00% , 50.00% and 30.00% respectively, and doxorubicin sensitive breast cells were respectively 30.00% ,45.00% , 15.00% and 10.00%, and the difference was statistically significant (P 〈0.01). ALDH1, Smo pro-tein and Glil protein in the breast cancer tissues were all higher than the ones in the tissues beside breast cancer(P 〈0.05). ALDH1, Smo protein and Glil protein in the doxorubicin resistance breast cells were all higher thanthe ones in doxorubicin sensitive breast cells (P 〈 0. 05 ). CONCLUSION The high expression of STAT3 andregulatory signaling pathway related protein of stem cell Hedgehog in the breast cancer may relate to the resistance of doxorubicin.
