摘 要:以UV为检测手段,通过对反应生成的6-氨基青霉烷酸(6-aminopenicillanic acid,6-APA)与对二甲氨基苯甲醛(paradimethylaminobenzaldehyde,PDAB)的显色反应,分别研究了在不同温度和pH值的水溶液中,在醇与NaH_2PO_4-Na_2HPO_4缓冲溶液配制成的混合液中固定化青霉素酰化酶(IPA)催化青霉素G(penicillin G,PG)酶活保留率随时间的变化.结果表明:在25~55℃,温度越高,青霉素催化能力越强;温度越低,酶活性保持率越高;在弱碱性条件下青霉素酰化酶催化能力强,在偏酸性或接近中性的条件下酶的保存稳定性较强;对多元醇,随着醇中羟基质量分数的减小,IPA的酶活保留率依次减小;对于一元醇,溶剂的极性与IPA活性呈反相关关系,即溶剂极性越强,IPA活性越低.This work investigated the catalytic activity variation trend of the immobilized penicillin acylase (IPA) under different operation conditions, i.e. temperature, pH and the mixture of alcohol/NaH2PO4 Na2HPO4 buffer solution, with the prolong of treating time. During this process, the chromogenic reac tion between 6 aminopenicillanic acid (6 APA), which was generated from the decomposition of penicillin G, and paradimethylaminobenzaldehyde (PDAB) was adopted as characterization method, UV Visible spectro photometer was conscripted as the characterization instrument. Results showed that in the temper ature range from 25 ℃ to 55 ℃ , the higher the temperature was, the stronger catalytic activity of IPA would be. While the lower the temperature, the more advantageous to keep the catalytic activity of IPA. The catalytic activity of IPA would be strong under the weak basic solutions, while the stability of IPA would be stronger under weak acidic or near neutral solutions. As far as the solution of multiple alcohol was concerned, The catalytic retention rate would successively decrease with the decrease of mass fraction of hy droxy in alcohol. As for the solution of mono alcohol, its polarity would exhibit a counte-correlation to the activi ty of IPA, namely the stronger the polarity of the solution was, the weaker the activity of IPA would be.
