摘  要:为薏苡叶绿体基因组生物信息的开发综合应用、品种分子鉴定及种质亲缘关系分析提供参考,根据Genebank数据库公布薏苡叶绿体基因组的DNA序列信息,利用生物信息学方法分析薏苡叶绿体基因组的SSR位点及分布特征,进而利用Primer 5.0软件设计SSR位点上下游250bp侧翼引物,并利用PCR扩增筛选SSR多态性引物,根据SSR扩增产物的多态性,利用UPGMA方法对14份薏苡种质材料进行聚类分析以确定其亲缘关系。结果表明:在全长140 745bp的薏苡叶绿体基因组DNA中,共搜寻到38个SSR位点,平均每3 835个碱基出现1个SSR位点。其中,在大单拷贝区(LSC)有30个位点,占所有标记数目的78.95%;小单拷贝区(SSC)有4个位点,占10.53%;2个反向重复区(IRa和IRb)各有2个位点,均占5.26%。单核苷酸重复的SSR位点有16个、二核苷酸重复5个、三核苷酸重复2个、四核苷酸重复9个、复合核苷酸重复4个、五核苷酸重复和六核苷酸重复均为1个,出现频率分别为42.11%、13.16%、5.26%、23.68%、10.53%、2.63%和2.63%。在筛选的38条引物中,有21条引物在14个薏苡种质材料中表现出多态性,占有效扩增引物的55.3%。聚类分析将14份薏苡种质材料分为两类,Y07号种质材料单独为Ⅰ类,其余13个种质材料聚为Ⅱ类。The SSR loci of adlay chloroplast gcnome and the distribution characteristics were analyzed by the bioinformaties according to DNA sequence information of adlay chloroplast genome published by Gencbank database and then the genetic relationship of 14 adlay germplasm resources was determined by UPGMA method according to the polymorphism of SSR amplification products to provide a reference for development and comprehensive utilization of chloroplast genome biological information, variety molecular identification and germplasm genetic relationship analysis in adlay. Result: 38 SSR loci arc detected from DNA of adlay chloroplast gcnomc with 140 745 bp and there is one SSR locus within 3835 bases averagely. 30 loci in LSC region account for 78.95% of total markers. 4 loci in SSC region account for 10. 53% of total markers. 2 loci in IRa region account for 5.26%; of total markers and 2 loci in IRh region accol,nt for 5.26% of tolal markers also. The number of SSR loci with mononucleotide repeat, dinucleotide repeat. trinucleotide repeal, tetrenucleotide repeal, compound neleotide repeal, pentanuclotide repeal and hexanucleotide repeal are 16, 5, 2, 9, 4.1 and 1 respeelively and their occurrence frequency is 42. 11%, 13.16%, 5. 26%, 23. 68%, 10. 53%, 2. 63% and 2. 63% separately. 21 primers of 38 screened primers performance polymorphism in 14 adlay germpIasm resources and account for 55. 3% of total effective amplification primers. 1..1 adlay germplasm resources are divided into two groups by a clustering analysis. Y07 germplasm material belongs to Group Ⅰ only and other 13 germplasm materials belong to Group Ⅱ.
