摘 要:目的探讨氢离子(H+)对碘酸钠(Na IO3)诱导的小鼠年龄相关性黄斑变性视网膜的保护作用及机制。方法选取60只8~10周龄SPF级健康C57BL/6雄性小鼠,随机分为对照组、Na IO3组、H+组,每组20只。H+组小鼠灌胃富含H+水(每只每天6 m L)预治疗7 d,对照组及Na IO3组小鼠灌胃相同剂量的自来水;Na IO3组、H+组小鼠按20 mg·kg-1剂量鼠尾静脉注射Na IO3溶液,对照组小鼠鼠尾静脉注射相同剂量的生理盐水;H+组小鼠继续灌胃富含H+水(每只每天6 m L)治疗5 d,对照组及Na IO3组小鼠给予相同剂量的自来水。然后对小鼠行眼底照相、OCT、视网膜电图、FFA检查,取小鼠眼球行HE染色,Western blot检测Caspase8表达情况。结果 Na IO3组小鼠视网膜厚度[(232.370±17.060)μm]与对照组[(316.265±5.314)μm]、H+组[(296.406±27.411)μm]间差异均有统计学意义(均为P<0.05),对照组与H+组之间差异无统计学意义(P=0.122)。Na IO3组a波振幅低于对照组和H+组,差异均有统计学意义(均为P<0.05),对照组a波振幅与H+组间差异亦有统计学意义(P=0.036)。对照组和H+组b波振幅均高于Na IO3组,差异均有统计学意义(均为P<0.05),对照组b波振幅与H+组间差异无统计学意义(P=0.121)。FFA显示,对照组小鼠无血管渗漏,Na IO3组小鼠的视网膜血管有大面积渗漏区,H+组小鼠视网膜渗漏面积较Na IO3组明显减少。HE染色结果表明,Na IO3组小鼠视网膜色素上皮层变薄甚至消失,并且出现大量的黑色沉积物,H+组小鼠视网膜黑色沉积物较Na IO3组减少。对照组(0.406±0.161)和H+组(0.543±0.302)视网膜中Caspase8相对表达量均明显低于Na IO3组(1.498±0.219),差异均有统计学意义(均为P<0.05)。对照组与H+组间差异无统计学意义(P=0.501)。结论 H+通过抑制Caspase8凋亡途径抑制Na IO3诱导的视网膜黄斑变性。Objective To investigate the protective effects of hydrogen ions (H +) on sodium iodate (NaIO 3)-induced age-related macular degeneration (AMD) in mice and its mechanisms.Methods Sixty 8-10 weeks old SPF healthy C57BL/6 male mice were randomly divided into control group,NaIO 3 group and H + group ( n =20).The mice in H + group were pretreated with H +-rich water (6 mL for one mouse every day) for 7 days,and mice in control group and NaIO 3 group were given the same dose of tap water.Mice in the NaIO 3 group and H + group were injected with NaIO 3 solution in tail vein with the dose of 20 mg·kg -1 ,while mice in control group were injected with the same dose of 9 g·L -1 physiological saline in tail vein.Mice in the H + group were treated with H +-rich water (6 mL for one mouse every day) for 5 days,and mice in the control group and NaIO 3 group were given the same dose of tap water.Then the mice were examined by fundus photography,OCT,ERG and FFA,and the eyeballs were extracted for HE staining.Western blot assay was used for the detection of Caspase8 expression.Results The retinal thickness of the NaIO 3 group [(232.370±7.060)μm] was significantly different from that of the control group [(316.265±5.314)μm] and H + group [(296.406±27.411)μm] (both P 〈0.05),but there was no significant difference between control group and H + group ( P =0.122).The amplitude of a wave in the NaIO 3 group was significantly lower than that in the control group and H + group,and the differences were statistically significant (both P 〈0.05).The amplitude of a wave in the control group was also significantly different from that in H + group ( P =0.036).The amplitude of b wave in the control group and H + group was significantly higher than that in the NaIO 3 group,and the differences were statistically significant (both P 〈0.05),but there was no significant difference in b wave amplitude between the control group and H + group ( P =0.121).
