摘  要:目的观察腺苷预处理的局灶性脑缺血再灌注损伤大鼠神经细胞线粒体功能。方法 30只雄性SD大鼠随机分为3组各10只,腺苷组大鼠制模前3 d腹腔注射腺苷注射液1.5 mg/kg(生理盐水稀释至2 m L),1次/d,连续3 d;假手术组和模型组给予等量生理盐水。腺苷组和模型组大鼠均制备大脑中动脉闭塞(MCAO)模型,术后2 h拔出栓线进行再灌注22 h;假手术组大鼠只暴露左侧颈总动脉而不结扎。比较各组神经细胞线粒体基质内Ca2+浓度、膜电位、Na+-K+-ATPase活性、三磷酸腺苷(ATP)含量。结果腺苷组、模型组、假手术组神经细胞线粒体基质Ca2+浓度分别为(157.07±25.12)、(233.12±24.58)、(106.85±17.21)nmol/L,膜电位分别为(4.36×104±171 8.285)、(3.90×104±1 777.309)、(5.39×104±2 516.563)相对荧光单位,Na+-K+-ATPase活性分别为(12.233±1.165)、(7.110±0.799)、(16.103±0.872)μmol Pi/(mg prot·h),ATP含量分别为(20.26±2.63)、(10.68±2.24)、(32.16±3.44)μmol/g prot,腺苷组、假手术组分别与模型组比较,P均<0.05。结论腺苷预处理能够改善大鼠局灶性脑缺血再灌注损伤后神经细胞线粒体功能。Objective To observe the effects of adenosine preconditioning on neuronal mitochondrial function after focal cerebral ischemia-reperfusion injury in rats. Methods Thirty male Sprague-Dawley rats were randomly divided into three groups with 10 in each. The rats in the adenosine group were intraperitoneally injected with adenosine 1. 5 mg/kg( normal saline diluted to 2 m L) at 3 d before model making,once a day,for 3 days; rats in the sham and model groups were given equal amounts of normal saline. The middle cerebral artery occlusion( MCAO) model was established in the adenosine group and the model group,and the suppository was extracted at 2 h after the operation and we continued to did reperfusion for 22 h; in the sham group we only exposed only the left common carotid artery without ligation. The mitochondrial Ca2 +concentration,membrane potential,Na^+-K^+-ATPase activity,and adenosine triphosphate( ATP) content were compared in each group. Results The neuronal mitochondrial matrix Ca2 +concentration in the adenosine group,model group,and sham group were( 157. 07 ± 25. 12),( 233. 12 ± 24. 58),and( 106. 85 ± 17. 21) nmol/L; the membrane potential was( 4. 36 × 10^4± 1 718. 285),( 3. 90 × 10^4± 1 777. 309),and( 5. 39 × 10^4± 2 516. 563) relative fluorescence units; Na^+-K^+-ATPase activity were( 12. 233 ± 1. 165),( 7. 110 ± 0. 799),and( 16. 103 ± 0. 872) μmol Pi/mg prot/h,respectively; the ATP content was( 20. 26 ± 2. 63),( 10. 68 ± 2. 24),and( 32. 165 ± 3. 44) μmol/g prot; significant difference was found between the adenosine and the model group,and between the sham group and the model group,all P〈0. 05. Conclusion Adenosine preconditioning can improve the function of mitochondria in brain after focal cerebral ischemia-reperfusion injury in rats.
