摘 要:目的探讨广藿香提取物对子宫内膜癌细胞相关耐药基因表达及紫杉醇耐药性的影响。方法选取对数生长期Ishikawa细胞株,给予8 nmol/L浓度的2,3,7,8-四氯苯二噁英稀释液诱导后,将其分为A组、B组、C组,分别加入0.1 mg/ml的紫杉醇、0.1 mg/ml的紫杉醇和0.1 mg/ml的广藿香提取物、0.1 mg/ml的紫杉醇和1.0 mg/ml的广藿香提取物,比较3组细胞的细胞色素P450 1B1(CYP1B1)、多药耐药(MDR-1)及多耐药相关蛋白(MRP)-1 mRNA及蛋白相对表达情况以及细胞生长抑制率。结果 A组、B组、C组细胞的CYP1B1、MDR-1、MRP-1 mRNA以及蛋白相对表达量依次降低(P<0.05)。A组、B组、C组细胞的生长抑制率亦依次升高(P<0.05)。结论广藿香提取物可以有效下调CYP1B1、MDR-1以及MRP-1 mRNA及其蛋白的表达量,从而抑制子宫内膜癌细胞对紫杉醇的耐药性,提高子宫内膜癌细胞的生长抑制率。Objective To investigate the effects of pogostemon cablin aqueous extract(PCAE) extract on related drug-resistance genes and paclitaxel resistance in endometrial carcinoma cells. Methods The Ishikeuvah cell lines in logarithmic growth phase were selected and were induced by 2,3,7,8- tetrachlorodibenzodioxin dilution with the concentration of 8 nmol/L.Then the cells were divided into group A,group B and group C.The cells in group A,group B and group C were added with paclitaxel(0.1 mg/ml),paclitaxel (0.1 mg/ml) +PCAE(0.1 mg/ml),and paclitaxel(0.1 mg/ml)+PCAE(1.0 mg/ml) respectively,The relative expressions of cytochrome P450 1B1(CYP1B1),multi-drug resistance-1(MDR-1) and multidrug resistance-associated protein(MRP-1) mRNAs and proteins ,and the inhibition rate of cell growth were compared among the three groups. Results The relative expression levels of CYP1B1,MDR-1 and MRP-1 mRNAs and proteins decreased in the order of group A,group B and group C( P 〈0.05). The inhibition rates of cell growth increased in the order of group A,group B and group C( P 〈0.05). Conclusion PCAE can effectively down-regulate the expression levels of CYP1B1,MDR-1 and MRP-1 mRNAs and proteins,thus inhibiting the resistance of endometrial cancer cells to paclitaxel and increasing the growth inhibition rate of endometrial cancer cells.
