长爪沙鼠全脑缺血再灌注损伤后半胱氨酰白三烯受体表达变化的研究

摘  要:目的研究半胱氨酰白三烯受体(CysLTR)在长爪沙鼠全脑缺血再灌注损伤后表达情况,并初步探讨CysLTR拮抗剂在脑缺血损伤治疗中的意义。方法采用结扎双侧颈总动脉结扎再灌注法制作110只长爪沙鼠全脑缺血再灌注损伤模型,并于再灌注24h、14d时对各组长爪沙鼠的神经症状和功能进行评分。全脑缺血10min后,分别再灌注3、6、12、24h及3、7、14d,使用Western blot法检测各个时点长爪沙鼠大脑皮层及海马中CysLT1R、CysLT2R的表达情况,使用免疫组化方法检测再灌注24h、14d这两个时点长爪沙鼠大脑皮层及海马中CysLT1R、CysLT2R的表达情况。结果与假手术组比较,全脑缺血模型再灌注24h、14d的神经症状评分增高和功能评分降低;全脑缺血10min后再灌注过程中,CysLT1R、CysLT2R表达呈动态变化,CysLT1R表达增高,出现2个峰(12h、14d),CysLT2R为1个峰(12h);在再灌注24h、14d,海马CA1区与皮层中CysLT1R、CysLT2R阳性细胞数量均明显增加。结论长爪沙鼠全脑缺血再灌注损伤后CysLT1R、CysLT2R蛋白表达并呈动态变化,并在皮层、海马区均有明显表达上调,提示CysLTR介导缺血后急性神经元损伤、亚急性期小胶质细胞的激活及慢性期星形胶质细胞的增生。Objective To investigate the expression of cysteinyl leukotrienes was receptors in gerbils brain after global common cerebral carotid ischemia/reperfusion(CI/R)followed injury.Methods 12,The and model of CI/R established in 110 gerbils by bilateral occlusion scores for 10 min by 3,6,24h-3,7,was 14d-reperfusion.After 24h-or 14d-reperfusion,neurological deficit were evaluated and the behavioral dysfunction cerebral assessed.and The expression of cysteinyl leukotrienes Western receptors(CysLT1R and CysLT1R and CysLT2R)in the CysLT2R positive homogenate cells of cortex and hippocampus were determined by blotting.The in cerebral cortex deficit hippocampus were observed by and immunohistochemistry.Results Compared to the sham CI/R group,the neurological 24hscores were The significantly expressions increased the behavioral CysLT2R in dysfunction gerbil was were aggravated in gerbils after or 14d-reperfusion.of CysLT1R and brain uponly 24h regulated one after at CI/R injury.CysLT1R expression The had two peaks at 12h and 14d,respectively,while CysLT2R expression were had peak 12h.CysLT1R and CysLT2R positive cells in cerebral cortex CysLT1R and and hippocampus CysLT2R increased markedly cortex at and 14d.Conclusion CI/R injury induces up-regulation of in gerbils These cerebral findings and that hippocampus,CysLTR the expression levels were dynamically and alterated during 3h-14d of reperfusion.global suggest may be involved in neuron damage gliosis(glial scar formation)following cerebral ischemic injury.
