摘 要:目的研究便秘型肠易激综合征(IBS-C)模型大鼠结肠黏膜下神经丛(SMP)中兴奋性和抑制性神经元的变化。方法用冰水灌胃法建立IBS-C大鼠模型,并设对照组。取肠道组织制作结肠SMP全层铺片标本;用免疫荧光多重染色技术检测SMP神经元及其活化状态;计数神经元的数量以及活化的特异性阳性神经元的总数和比例。结果IBS-C组(n=7)大鼠粪便粒数减少且含水量下降,肠道传输减慢。结肠SMP每高倍视野神经元总数和活化神经元总数均显著增多(P<0.05);活化的胆碱乙酰转移酶免疫反应性(IR)阳性神经元比例和活化的血管活性肠肽IR阳性神经元比例则显著降低(P<0.05);活化的一氧化氮合酚-免疫反应性(NOS-IR)阳性神经元比例显著增高(P<0.05)。结论结肠SMP的变化可能在IBS-C的发病中起重要作用。Objective To investigate the changes of excitatory and inhibitory neurons in submucosal plexus( SMP)of colon in a rat model of IBS-C. Methods The rat model of IBS-C was established by intragastric perfusion with cold water. Whole-thickness colon specimens were collected to prepare submucosal layer with intact SMP. Multiple immunofluorescence staining technique was used for detect the total numbers of SMP neurons and ganglia,the numbers of the SMP neurons expressing specific neurochemical markers( Ch AT,VIP,NOS),and the activation status of SMP neurons. Results Compared with control group,the intestinal transmission rate of model rats was slower,and the number of fecal granules and water content of feces were less than that of controls. In the colon SMP of rat with IBS-C( n = 7),the total number of SMP neurons/high power field( HPF) and the total number of activated neuron/HPF increased significantly( P〈0. 05); the percentages of activated Ch AT-IR neurons and activated VIPIR neurons decreased significantly, and the percentage of activated NOS-IR neurons increased significantly.Conclusions The change of colonic SMP may play an important role in the pathogenesis of IBS-C.
