灵菌红素微孔板发酵条件优化

摘　　要：为建立灵菌红素高产突变株快速培养和检测方法,比较了酶标仪与分光光度计检测灵菌红素产量之间的一致性及摇瓶、24孔板、48孔板发酵灵菌红素产量之间的关联性,结果表明:酶标仪与分光光度法之间具有很好的一致性,24株突变株产灵菌红素孔板发酵液两种测定方法的相关系数R^2=0.9788。摇瓶灵菌红素产量要高于24孔板,24孔板产量高于48孔板,而且三者具有很好的关联性。通过单因素试验对24孔板发酵产灵菌红素条件进行了优化,结果表明:当接种量6%、装液量2mL、转速200r/min时,24孔板发酵产灵菌红素产量最高,达到了0.948g/L,较最初产量的0.833g/L提高了13.8%。In order to establish a rapid culture and detection method for high yield mutant strains of prodigiosin,this experiment first compared the consistency between the microplate reader and the spectrophotometer for detection of the content of the prodigiosin, and the correlation in the fermentation of prodigiosin from shaking flask,24-hole and 48-hole plates. The results showed that there have a good consistency between the microplate reader and the spectrophotometric method, the correlation coefficient of these two methods could reach 0.9788 by determination the prodigiosin yield of 24 mutant strains fermented in 24-hole plates. The yield of the prodigiosin was higher than that of the 24-hole plate, and the output of the 24-hole plate was much higher than that of the 48-hole plate, and these three also had a good relevance. Secondly, the 24-hole plate fermentation conditions were optimized by single factor. The results showed that the optimumal conditions were as follows： the inoculation amount 6%, the amount of liquid 2 mL, and the speed 200 r/min respectively. Under these conditions, the prodigiosin yield of 24 hole plate fermentation reached 0.948 g/L, which was 13.8% higher than that of the initial yield0.833 g/L.
