凋亡抑制因子Livin和Survivin在乳腺癌中的表达及临床意义

摘　　要：目的探讨凋亡抑制因子Livin和Survivin在乳腺癌中的表达及临床意义。方法选择56例女性乳腺癌患者手术切除组织为研究样本,通过免疫组化技术和RT-PCR技术检测Livin和Survivin蛋白及mRNA在乳腺癌中的表达情况。结果 56例乳腺癌组织中Survivin mRNA和Livin mRNA分别有40例和34例阳性表达,显著高于癌旁组织(P<0.05)。免疫组化结果中,56例乳腺癌样本中分别有38例和31例染色,癌旁组织中则分别有5例和3例染色,两者差异具有统计学意义(P<0.05)。Livin mRNA表达与患者的年龄、肿瘤大小、TNM分期、淋巴结转移、雌激素受体、Her2/neu均无关;Survivin mRNA的表达与淋巴结转移显著相关(P<0.05)。Survivin蛋白和Livin蛋白在乳腺癌组织中表达无相关性(rs=-0.198,P=0.117)。结论肿瘤细胞凋亡抑制因子Livin和Survivin上调与乳腺癌的发生和发展密切相关,Survivin高表达与患者淋巴结转移显著相关,具有临床诊断价值,Livin和Survivin可以作为治疗乳腺癌的靶基因。Objective：To investigate the expression and clinical significance of apoptosis inhibitory factor Livin and Survivin in breast cancer.Methods：A total of 56 women with breast cancer were selected as the study group.The expressions of Livin and Survivin protein and mRNA were detected by immunohistochemistry and RT-PCR.Results：There were 40 cases of Survivin mRNA and Livin mRNA in 56 cases of breast cancer,and 34 cases were positive,which was significantly higher than that of para cancerous tissue（P0.05）.Among the 56 cases of breast cancer,38 cases and 31 cases were stained by immunohistochemistry.There were 5 cases and 3 cases in the adjacent tissues respectively.The difference was statistically significant（P0.05）.The expression of Livin mRNA was not related to age,tumor size,TNM stage,lymph node metastasis,estrogen receptor or Her2/neu.The expression of Survivin mRNA was significantly correlated with lymph node metastasis（P0.05）.There was no correlation between the expressions of Survivin protein and Livin protein in breast cancer tissues（rs=-0.198,P=0.117）.Conclusion：The up-regulation of Livin and Survivin in tumor cells is closely related to the occurrence and development of breast cancer.The high expression of Survivin is significantly correlated with lymph node metastasis in patients,which has clinical diagnostic value.Livin and Survivin can be used as target genes in the treatment of breast cancer.
