摘  要:目的:探讨楝酰胺衍生物Aglaroxin C对小鼠H22肝癌皮下移植瘤的影响及在实验剂量下对小鼠各脏器有无影响。方法:建立BALB/C小鼠H22腋下实体瘤模型,并随机分为Ⅰ组(尾静脉给药对照组)、Ⅱ组(尾静脉给药组,0.1 mg每只)、Ⅲ组(瘤内给药对照组)、Ⅳ组(瘤内给药组,0.05 mg每只)、Ⅴ组(瘤内给药组,0.1 mg每只),每组各5只。隔天给药(Aglaroxin C)一次,共4次。每日测量小鼠体重。末次给药24小时后切除肿瘤及各组小鼠主要脏器并称重,计算抑瘤率、脏器系数。用各组肿瘤及各脏器制作病理切片,苏木精-伊红染色(hematoxylin-eosin staining,HE染色),并在光镜下观察其细胞形态学变化。结果:Ⅰ组、Ⅱ组、Ⅲ组、Ⅳ组、Ⅴ组的瘤体质量分别为(2.11±0.50)g、(1.59±0.14)g、(2.30±0.64)g、(1.66±0.12)g、(1.06±0.14)g,Ⅱ组瘤体质量明显小于Ⅰ组(P<0.01),Ⅲ组、Ⅳ组、Ⅴ组比较差异有统计学意义(P<0.01)。Ⅴ组的抑瘤率最高达53.91%。显微镜下观察肿瘤病理切片,给药组坏死肿瘤细胞明显增多,对照组肿瘤细胞坏死轻微;各组小鼠体质量组间变化无明显统计学意义(P>0.05);显微镜下观察各脏器病理切片,各给药组与对照组相比,无明显异常;脏器系数各剂量组与对照组组间比较,差异无统计学意义(P>0.05)。结论:Aglaroxin C对小鼠H22肝癌皮下移植瘤的生长有抑制作用,在实验剂量下对小鼠各脏器无明显影响。Objective: To investigate the anti-tumor effects of Aglaroxin C in H22 tumor-bearing mice and its influence on mice organs at the experimental dose.Methods: The solid tumor model was established by subcutaneously transplanting H22 cells into BALB/C mice.The mice were subsequently divided into five groups as follows: Group Ⅰ( tail vein administration of physiological saline,negative control group),group Ⅱ( tail vein administration of 0.1 mg Aglaroxin C),group Ⅲ( intratumoral administration of physiological saline,negative control group),group Ⅳ( intratumoral administration of 0.05 mg Aglaroxin C),group Ⅴ( intratumoral administration of 0.1 mg Aglaroxin C).Each group had 5 mice.The mice in other groups were given drugs at indicated time point for a total of 4 times.The body weight of mice was measured daily.24 hours after the last drug injection,all mice were sacrificed.Main organs and tumor tissues were collected and weighed.The tumor inhibition rate and organ coefficient were calculated.Results: The mean tumor weights( g) in the different groups were : Ⅰ( 2.11 ± 0.50),Ⅱ( 1.59 ± 0.14),Ⅲ( 2.30 ±0.64),Ⅳ( 1.66 ± 0.12),Ⅴ( 1.06 ± 0.14)( in grams).The tumor weight from groupⅡ was significantly less than that of groupⅠ( P 0.01).There was also significant differences in tumor weight between group Ⅲ,Ⅳ and Ⅴ( P 0.01).The highest inhibition rate was noticed in group Ⅴ( 53.91%).Necrotic tumor cells in Aglaroxin C treated groups increased significantly.By contrast,body weight changes between groups were not statistically significant( P 0.05).Micromorphological changes of organs showed no difference between the control and experimental groups.Furthermore,there was no significant difference in organ coefficient between the control and experimental groups( P 0.05).Conclusion: Aglaroxin C potently inhibited the proliferation of mice hepatoma H22 cells in the mice tumor model.At effective antitumor concentrations,Agl
