摘 要:目的优选龙葵多糖脱蛋白的最佳工艺。方法以多糖保留率、蛋白脱除率为指标,采用正交试验确定Sevag法、三氯乙酸法(TCA法)、酶法的最佳工艺条件,并进行比较。结果三氯乙酸法(TCA法)为最佳脱蛋白方法,最佳工艺条件为用3%的三氯乙酸溶液调节pH至2.5,萃取时间20分钟,萃取次数1次,此时蛋白脱除率87.118%,多糖保留率88.983%。结论 TCA法对龙葵多糖具有较好的脱蛋白效果,且操作简便,工艺稳定,多糖保留率高,可在龙葵多糖的分离纯化中推广使用。Objective To optimize deproteinization process of solauum nigrum polysaccharide. Methods Retention rate of polysaccharide and clearance rate of protein were used as indexes, orthogonaltests were used to determine the optimum conditions of sevag method, trichloroacetic acid method and enzyme method, then the three method were compared. Results TCA method was the best method of deproteinization. The optimum process conditions were as follows: 3% trichloroacetic acid solution was used to adjust pH to 2. S, extraction time was 20 min, extraction frequency was 1 times. Then the clearance rate of protein was 87. 118% , the retention rate of polysaccharide was 88. 983%. Conclusion TCA method has good effect of deproteinization of solanum nigrum polysaccharide, and its operation is simple, the process is stable and the retention rate of polysaccharide is high. It can be used in the separation and purification of the polysaceharide in solannum nigrum.
