摘 要:目的观察GDF11和维生素D对UMR106细胞增殖及成骨相关转录因子Runx2 mRNA、Osterix mRNA表达的影响。方法培养UMR106细胞,通过MTT法、ALP活性检测及q-PCR技术测定GDF11(50、100ng/m L)、维生素D(10-6mol/l)及联合应用对细胞增殖、Runx2 mRNA和Osterix mRNA的表达。结果 (1)维生素D(24 h)和GDF11均能促进细胞增殖,维生素D与低浓度GDF11联合应用24、48、72 h,对细胞增殖具有协同效应,而与高浓度GDF11联合应用,则为拮抗效应;(2)与对照相比,除维生素D组,各组ALP活性明显增加;(3)与对照相比,除维生素D组,各组细胞Runx2 mRNA表达均明显提高,Osterix mRNA只在24、48 h高表达。结论 (1)GDF11可通过提高Runx2 mRNA和Osterix mRNA的表达,促进细胞增殖及提高ALP活性;(2)维生素D仅在短时间内促进细胞增殖,可能并非通过Runx2信号通路发挥作用;(3)维生素D影响GDF11促进细胞增殖的作用。Objectiv To investigate the effects GDF11 and Vitamin D on the proliferation of UMR106 cells and the expression of Runx2 mRNA and Osterix mRNA. Methods The UMR106 cells were co-cultured with GDF11(50, 100 ng/m L) and Vitamin D(10-6 mol/l). The proliferation of cells, ALP activity, Runx2 mRNA and Osterix mRNA expression was detected by MTT assay, ALP activity assay and q-PCR at different time points, respectively. The results showed that: Results(1) Both Vitamin D(24 h) and GDF11 promoted cell proliferation. Administration of Vitamin D and low concentration of GDF11 to cultured cells on 24,48 and 72 h had synergistic effect on cell proliferation, Vitamin D and high concentration of GDF11 had antagonistic effect;(2) Compared with the control, the activity of ALP was significantly increased in each group except for the Vitamin D;(3) Compared with the control,the expression of Runx2 mRNA was significantly increased in each group and the expression of Osterix mRNA was only increased on 24 and 48 h, expect the Vitamin D. Conclusion It can be concluded that GDF11 promotes the proliferation of cell by improving the expression of Runx2 mRNA and Osterix mRNA and enhances ALP activity; Vitamin D promotes cell proliferation in a short period time and does not play a role through the Runx2 pathway; Vitamin D affects the role of GDF11 in promoting cell proliferation.
