摘 要:目的:探讨不同大小的正畸力对大鼠磨牙牙根发育的影响。方法:取5周龄健康雄性SD大鼠96只,随机分为3个实验组和1个空白对照组(n=24)。对3个实验组大鼠建立正畸模型后,分别对其施加50(A组)、75(B组)、100 g(C组)的正畸力;空白对照组大鼠不做任何处理。分别于正畸加力后1、3、7、14 d各时间点每组随机处死6只大鼠,取其左侧上颌骨并拍摄CBCT,比较大鼠第一磨牙近中根的长度;然后对颌骨进行连续切片后,进行HE染色及BSP、MMP-9免疫组化染色。结果:CBCT测量结果显示,与空白对照组相比,第一磨牙近中根长度B组在加力后14 d时及C组在加力后7、14 d时均有统计学差异(P<0.05)。免疫组化染色结果显示,与空白对照组相比,B组在加力后14 d时BSP表达下调(P<0.05),而在7 d时MMP-9表达开始上调(P<0.05);C组在加力7 d时BSP表达下调、14 d时达到峰值(P<0.05),从加力3 d时,MMP-9随着加力时间的延长其表达水平逐渐升高(P<0.05)。结论:BSP及MMP-9的表达均与正畸力值大小及加力时间相关,其中100 g的正畸力对大鼠牙根发育影响较大。AIM: To investigate the effects of orthodontic force on root developmenl of rat molars. METH- ODS : 5 - week - old healthy male SD rats were used for the establishment of orthodontic models and treated with the or- thodontic force of 50 g(group A), 75 g( group B), 100 g( group C) and 0 g(the control, goup D) respectively. The rats were sacrificed at 1 d,3 d,7 d and 14 d respectively(n =6). After CBCT photography, HE staining, bone sialoprotein (BSP) and MMP-9 immunohistochenfical staining were performed. RESULTS : CBCT results showed that the root length of the first molars of group B was longer thin1 that of group D(P 〈0.05) at 7 and 14 days. BSP and MMP -9 analysis results showed in group B, the expression of BSP was down - regulated ( P 〈 0.05 ) at 14 days after treatment, while the expression of MMP-9 was up-regulated(P 〈0.05) at 7 days. In group C, the expression of BSP was down -regulated (P〈0.05) on the 7th day after adding force until 14 days, but the expression of MMP -9 was up -regulated (P 〈0.05) at the 3rd day. The gray value of group C was significanlly higher than that of the control group(P 〈0.05). CONCLUSION : 100 g orthodontic force have a greater effect on the root development of rats in a time - dependent manner.
