摘 要:目的探讨百里醌(TQ)是否增强吉非替尼对EGFR-L858R/T790M突变的H1975细胞的毒性作用。方法人类非小细胞肺癌(NSCLC)细胞系95-D、A549、SK-MES-1、H1299、H1975,正常肝脏细胞系HL7702和正常气道上皮细胞系BEAS-2B,按1×10~4/孔,接种于96孔板,予20μmol/L、40μmol/L、60μmol/L、80μmol/L、100μmol/L TQ培养24 h,MTT测细胞存活率,筛选对TQ敏感的细胞系。筛选细胞后比较TQ、顺铂、吉非替尼三种药物72 h的细胞存活率,评价TQ联合顺铂,或者TQ联合吉非替尼对筛选细胞系的毒性增敏作用。结果 TQ对95-D和H1975细胞系比较敏感;培养72 h后,TQ对95-D细胞的毒性作用不优于顺铂和吉非替尼,但在H1975细胞中TQ毒性优于顺铂,但劣于吉非替尼,TQ组和TQ+吉非替尼组的细胞存活率比较差异无统计学意义(P>0.05);Chou-Talalay公式计算表明TQ和吉非替尼既无相加也无协同作用。结论 TQ联合吉非替尼治疗不增加EGFR-L858R/T790M突变H1975细胞系的细胞毒性作用。Objective To investigate whether Thymoquinone(TQ) enhances the cytotoxicity of Gefitinib on EGFR-L858 R/T790 M mutant H1975 cells. Methods Human non-small cell lung cancer(NSCLC) cell lines 95-D,A549, SK-MES-1, H1299, H1975, normal human liver cell line HL7702 and normal human bronchial epithelial cell line BEAS-2 B were inoculated in 96-well plates at 1×10^4/well, and cultured for 24 hours after administration of 20 μmol/L,40 μ mol/L, 60 μ mol/L, 80 μ mol/L and 100 μ mol/L TQ, respectively. Cell viability was measured by MTT, and cell lines sensitive to TQ were screened. Then the cell viability was compared after 72 h incubation with TQ, Cisplatin and Gefitinib respectively. The sensitization effects were evaluated on screened cell lines with TQ plus Cisplatin or TQ plus Gefitinib. Results TQ was sensitive to 95-D and H1975 cell lines. After 72 h of incubation, the toxic effect of TQ on 95-D cells was not superior to that of Cisplatin and Gefitinib. TQ toxicity was higher than Cisplatin in H1975 cells but inferior to Gefitinib, and there was no statistically significant difference in cell viability between the TQ group and the TQ plus Gefitinib group(P0.05). The calculation of Chou-talalay formula showed that TQ and Gefitinib had neither additive nor synergistic effect. Conclusion TQ combined with Gefitinib treatment dose not increase the cytotoxicity of EGFR-L858 R/T790 M mutant H1975 cell line.
