摘  要:目的测定糖尿病大鼠Bcl-2,Bax在视网膜的表达以及羟苯磺酸钙、牛蒡苷、槲皮素、葛根素、黄芩苷、水飞蓟素、柴胡皂苷、人参皂苷8种药物治疗糖尿病视网膜病变(DR)的作用。方法 7周龄雄性SD大鼠300只,随机取270只给予适应性喂养1周后,单次腹腔注射70mg/kg链脲佐菌素(STZ)造模;另30只作为正常对照组给予常规饲料,仅向腹腔注射同等量的枸橼酸缓冲液。将造模成功的270只大鼠随机取240只,平均分为8组,分别每日给予羟苯磺酸钙150mg/kg、牛蒡苷150mg/kg、槲皮素150mg/kg、葛根素150mg/kg、黄芩苷150mg/kg、水飞蓟素150mg/kg、柴胡皂苷150mg/kg、人参皂苷150mg/kg灌胃;另30只作为糖尿病对照组每日给予10ml/kg生理盐水。期间测定大鼠体重、血糖、尿白蛋白。32周后处死大鼠,取大鼠左眼ABC法免疫组织化学染色以观察Bcl-2,Bax阳性信号,TUNEL法检测凋亡细胞及计算细胞凋亡率。取大鼠右眼视网膜western blot法测各抗体条带灰度值,计算各组Bcl-2,Bax条带与β-肌动蛋白条带灰度比值,为Bcl-2,Bax蛋白的相对表达水平,比较Bcl-2/Bax值。结果羟苯磺酸钙组、黄芩苷组、牛蒡苷组、槲皮素组、葛根素组、水飞蓟素组、柴胡皂苷组、人参皂苷组的细胞凋亡率均低于糖尿病对照组(P<0.05)。羟苯磺酸钙组、黄芩苷组、牛蒡苷组、槲皮素组、葛根素组、水飞蓟素组、柴胡皂苷组、人参皂苷组Bcl-2/Bax与糖尿病对照组相比增加(P<0.05)。结论 Bcl-2,Bax参与大鼠糖尿病视网膜病变的发展过程,羟苯磺酸钙、黄芩苷、牛蒡苷、槲皮素、葛根素、水飞蓟素、柴胡皂苷、人参皂苷通过增加Bcl-2/Bax的比值抑制视网膜神经节细胞凋亡,对糖尿病视网膜病变起治疗作用。Objective To determine the expression of Bcl-2 and Bax in the retina of diabetic rats and the effect of eight kinds of drugs,Calcium Dobesilate,Arctiin,Quercetin,Puerarin,Baicalin,Silymarin,Saikoside and Ginsenoside on diabetic retinopathy. Methods A total of 270 rats were randomly selected from 300 male SD rats that aged 7 weeks and given adaptive feeding for 1 week,then underwent single intraperitoneal injection of 70 mg/kg Streptozotocin( STZ) for modeling; the other 30 rats were given routine feed only as a normal control group,and the same amount of citric acide buffer was injected into the abdomen. The 240 rats were randomly selected form the successfully modeled diabetic rats,and divided into 8 groups,which were given Calcium Dobesilate,Arctiin,Quercetin,Puerarin,Baicalin,Silymarin,Saikoside and Ginsenoside( each for 150 mg·kg^(-1)·d^(-1) respectively) to irrigate the stomach. The other 30 rats were given 10 ml·kg^(-1)·d^(-1) normal saline as diabetic control group. The weight,blood sugar and urine albumin of rats were determined. After 32 weeks,the rats were executed; then the left eyes were used for analyzing apoptosis rate by TUNEL method and for observing the positive signal of Bcl-2 and Bax by ABC immunohistochemical stain method. The retina of the right eyes were used for analyzing the Bcl-2 and Bax expression by western blot method. and Bcl-2/Bax were to known. Results The rates of apoptosis in Calcium Dobesilate group,Arctiin group,Quercetin group,Puerarin group,Baicalin group,Silymarin group,Saikoside group and Ginsenoside group were lower than those in diabetic control group( P〈0. 05).The value of Bcl-2/Bax in Calcium Dobesilate group,Arctiin group,Quercetin group,Puerarin group,Baicalin group,Silymarin group,Saikoside group and Ginsenoside group were higher than those in the diabetic control group( P〈0. 05). Conclusion Calcium Dobesilate,Arctiin,Quercetin,Puerarin,Baicalin,Silymarin,Saikoside and Ginsenoside can inhibit cell apoptosis by increasing the rat
