摘  要:目的以宫颈癌U14细胞小鼠皮下移植瘤模型为研究对象,探讨二甲双胍联合卡铂对宫颈癌U14小鼠移植瘤模型肿瘤的体内抗肿瘤作用。方法建立宫颈癌U14细胞小鼠皮下肿瘤模型,将其分为生理盐水对照组、二甲双胍组、卡铂组、二甲双胍联合卡铂组,每组各9只;二甲双胍100 mg/kg/day注入小鼠腹腔内,共18天,卡铂以50mg/kg注入小鼠腹腔内,共1次,对照组腹腔内注入等量的生理盐水。给药后开始记录荷瘤小鼠的生存状态,且每2天测定其肿瘤体积及小鼠体重,18天后处死小鼠,并对其肿瘤体积、小鼠体重、平均生存期进行比较,同时留取各组小鼠肿瘤组织观察其病理组织学变化。结果二甲双胍联合卡铂给药组能显著抑制肿瘤生长,与其他3组对比具有极显著性差异(P<0.01);实验周期内,各组小鼠体重均有所增长,但组间无明显差异,证实各组给药剂量下均无明显毒性反应;HE染色病理结果显示,与其他各组比较,联合给药组肿瘤组织多见局灶性坏死,细胞核深染,呈褐色,细胞核固缩、溶解、碎裂,染色质部分红染。结论体内实验研究证实,二甲双胍联合卡铂能显著抑制宫颈癌U14细胞株小鼠皮下移植瘤的生长,二甲双胍能够增强卡铂的化疗敏感性。Objective The anti-tumor effect of metformin combined with carboplatin was studied with the model of subcutaneous transplantation tumor in mice with cervical cancer U14 cells in vivo.Methods To establish the subcutaneous xenograft tumor model of cervical cancer U14 cell in mice,and the mice were divided into control group,metformin group,carboplatin group,metformin group combined with carboplatin group,9 mice in each group;Metformin was injected into the abdominal cavity of mice with 100 mg/kg/day for 18 days,and carboplatin was injected into the abdominal cavity of mice with 50 mg/kg for 1 time.The abdominal cavity of the control group was injected with the same amount of sterile saline.As the experiment began,we recorded the survival state of tumor-burdened mice,and monitoring the tumor volume and weight of mice every 2 days.At the end of the experiment,after mice were killed,We compared the differences of tumor size,body weight between the groups,and at the same time the histopathological changes of tumor tissues in mice were observed.Results Metformin combined with carboplatin could significantly inhibit tumor growth and improve the life cycle of tumor-bearing mice,which was significantly different from the other three groups(P0.01).During the experimental period,the weight of mice in each group increased,but there was no significant difference between groups.Compared with other groups,the HE stained section of tumor tissue in joint group show that cells are loose,seeing more focal necrosis,and the nuclei are hyperchromatic,brownish,nucleus pycnosis,dissolution,fracture,chromatin is dyed red.Conclusion Metformin can inhibit the growth of subcutaneous xenograft tumors in mice with cervical cancer U14 cell In vivo studies and at the same time Metformin can enhance the sensitivity of carboplatin to chemotherapy.
