摘 要:目的探讨缬草提取物对睡眠障碍阿尔茨海默病(SDAD)模型大鼠学习记忆与抗氧化能力的影响。方法先腹腔注射D-半乳糖促衰老,再剥夺衰老大鼠睡眠至睡眠障碍,最后衰老睡眠障碍大鼠侧脑室一次性注射β淀粉样蛋白(Aβ)1~40制备SDAD模型。选用SD大鼠,随机分13组,分别为空白组,SDAD模型组,缬草醇提物脱脂高、中剂量组,水洗脱物高、中剂量组,20%乙醇洗脱物高、中剂量组,40%乙醇洗脱物高、中剂量组,70%乙醇洗脱物高、中剂量组,阳性药多奈哌齐组。各给药组于术后3 d开始灌胃给药6 d。采用Morris水迷宫测试大鼠学习记忆能力,自主活动装置检测大鼠自主活动次数,并测定大鼠血清中过氧化氢酶(CAT)、总抗氧化能力(T-AOC)、乙酰胆碱酯酶(AChE)的含量和超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的活力。结果与模型组比较,给药各组能明显缩短潜伏期、增加过台次数、在第Ⅳ象限逗留时间延长,自主活动次数恢复,并能使大鼠血清中的CAT、T-AOC含量升高、降低AChE活力,可提高SOD、GSH-Px的活性,降低脂质过氧化物MDA含量(P<0.05,P<0.01)。结论缬草提取物(尤其是缬草醇提物脱脂组、20%乙醇洗脱物组和70%乙醇洗脱物组)对SDAD模型大鼠学习记忆和自主活动有明显的改善作用,其机制可能与增强T-AOC的活力、降低AChE含量及提高SDAD大鼠的抗氧化作用有关。Objective To screen effect of extracts from Valeriana officinalis on spatial learning memory and antioxidant capacity in rat model of sleep disorder Alzheimer's disease( SDAD). Methods Firstly,the rats were injected with galactose into abdominal cavity to senescent,secondly the aging rats sleep was deprived to sleep disorder,lastly the sleep disorder rats were intracerebroventricular injected with Aβ1-40 once to induce full SDAD model. The experimental rats were divided into normal control,model,Aricept,high and middle dose of total extract of Valeriana officinalis,the water elution high and middle dose of Valeriana officinalis,the 20% ethanol elution high and middle dose of Valeriana officinalis,the 40% ethanol elution high and middle dose of Valeriana officinalis,the 70% ethanol elution high and middle dose of Valeriana officinalis groups. Morris water maze was applied to test the spatial learning memory ability of each group rats.Meanwhile,plasma contents of CAT,T-AOC,AChE,SOD,MDA and GSH-PX were detected. Results Compared with those of model group,the incubating periods in drug delivery groups were reduced,through the platform times were increased,in the fourth quadrant stay time were extended,serum contents of CAT and T-AOC were increased,serum vitality of AChE were reduced,serum activities of SOD and GSH-PX were increased,serum content of MDA were decreased( P 0. 05,P 0. 01). Conclusions The extract of Valeriana officinalis( especially the total extract of Valeriana officinalis,the 20% ethanol elution group and the 70% ethanol elution group) has obvious improvement effect on spatial learning memory in SDAD model rats,its mechanism might be associated with the enhancement of T-AOC activity and lower AChE content,effectively inhibit the lipid peroxide and remove excess free radicals in the body,significantly improve the antioxidant capacity of SDAD rats.
