摘  要:目的探讨非酒精性脂肪性肝病(NAFLD)小鼠肝组织WD重复结构域35(WDR35)mRNA及蛋白的表达情况及其意义。方法选择健康雄性昆明小鼠36只,随机分为对照组和模型组,各18只,分别给予普通饲料、蛋氨酸-胆碱缺乏饲料喂养,分别于4周、8周和12周后各组取6只小鼠处死,检测血清ALT、AST、白细胞介素-6(IL-6)水平,光镜下观察肝脏组织病理学变化并计算NAFLD活动度积分(NAS),检测肝组织WDR35 mRNA及蛋白表达水平。结果镜下可见模型组肝组织出现不同程度脂肪沉积、炎症细胞浸润伴有空泡细胞形成,提示造模成功;各时间点模型组NAS均高于对照组,且随时间延长而升高(P<0.05)。各时间点模型组血清ALT、AST、IL-6水平及肝组织WDR35 mRNA相对表达水平高于对照组(均P<0.05),肝组织WDR35蛋白表达水平亦较对照组增加。结论 NAFLD小鼠肝脏组织中WDR35 mRNA以及蛋白的表达均增加,WDR35可能因与炎症因子相互作用而在NAFLD的发生中发挥重要作用。Objective To investigate the expressions and significance of WD repeat domain 35(WDR35) mRNA and protein in liver tissues of mice with nonalcoholic fatty liver disease(NAFLD). Methods Thirty-six healthy male Kunming mice were randomly divided into control group and model group,with 18 mice in each group. The control group and model group were fed with ordinary diet and methionine-choline deficient diet respectively. At 4 weeks,8 weeks and 12 weeks after modeling,6 mice of each group were killed separately.Serum levels of ALT,AST and interleukin-6(IL-6) were measured. The pathologic changes of hepatic tissues were observed under microscope and NAFLD activity score(NAS) was assessed. The expression levels of WDR35 mRNA and protein in liver tissues were detected. Results Fat deposition with various degrees,inflammatory cell infiltration,and physaliferous cells were observed in the liver tissues of the model group under microscope,which indicated successful modeling. The NAS of the model group was higher than that of the control group at each time point,and increased by the time(P〈0. 05). At each time point,the serum ALT,AST and IL-6 levels,relative expression level of WDR35 mRNA in liver tissues were higher(all P〈0. 05) and the expression level of WDR35 protein also increased in liver tissues in the model group compared to the controls. Conclusion The expressions of WDR35 mRNA and protein increase in liver tissues of NAFLD mice,and WDR35 may play an important role in the incidence of NAFLD by interaction with inflammatory factors.
