摘 要:目的探究干预富含AT序列的特异结合蛋白1(STAB1)基因表达对甲状腺癌TPC-1细胞侵袭、迁移的影响及其分子机制。方法脂质体介导下沉默甲状腺癌TPC-1细胞中STAB1基因的表达,蛋白质印迹法(Western blot)检测转染后细胞中STAB1蛋白的表达;细胞创伤愈合实验检测细胞的愈合率,Transwell小室法测定细胞的侵袭、迁移能力,Western blot检测细胞β-连环蛋白(β-catenin)、磷酸化β-catenin(p-β-catenin)、细胞周期蛋白D1(Cyclin D1)的水平。结果与正常对照组相比,STAB1干扰组细胞中STAB1蛋白表达显著下调(P<0.05),细胞愈合率及侵袭、迁移能力显著降低(P<0.05),β-catenin、Cyclin D1蛋白水平显著降低(P<0.05),p-β-catenin蛋白水平显著升高(P<0.05)。结论干预STAB1表达可抑制甲状腺癌TPC-1细胞的侵袭和迁移,其作用机制与Wnt/β-catenin信号通路密切相关。Objective To investigate the effects and possible molecular mechanism of interfering of Special AT rich sequence blinding protein 1 (STAB 1) gene expression on invasion and migration of thyroid carcinoma TPC- 1 cells. Methods Thyroid cancer TPC- 1 cells as the research object, the expression of STAB1 gene was silenced by liposome. The expression of STAB1 protein was analyzed by Western blot. The cell healing rate was detected by wound healing test. The invasion and migration of cells were detected by Transwell assay. The levels of β-catenin, p-β-catenin and Cyclin D1 in the cells were detected by Western blot. Results Compared with the normal control group, STAB1 protein expression in STAB 1 interfected group was significantly decreased (P 〈 0.05), and the healing rate, invasion and migration were sig- nificantly decreased (P 〈 0.05). The protein levels of β-catenin and Cyclin D1 were significantly down-regulated in STAB 1 interfected group (P 〈 0.05), while the protein expression of p-β-catenin was significantly up-regulated (P 〈 0.05). Conclusion The intervention of STAB 1 expression could inhibit the invasion and migration of TPC-1 cells, and its function may be closely related to the Wnt/β-catenin signaling pathway.
