摘 要:目的葡萄糖无氧糖酵解被认为是肿瘤细胞的重要能量代谢方式之一,但近来研究发现,谷氨酰胺也是肿瘤细胞赖以生存的重要营养物质。肿瘤细胞在内外因素驱动下发生谷氨酰胺代谢重编程,可从细胞外摄取谷氨酰胺。SLC1A5是细胞吸收外源性谷氨酰胺最主要的转运蛋白。细胞通过SLC1A5来调控谷氨酰胺转运进入细胞内进而为细胞代谢提供能量来源。SLC1A5在多种肿瘤中已有报道,但在胶质瘤中尚未见有相关研究报道。本研究探讨SLC1A5在胶质瘤中表达的临床病理学意义及在体外胶质瘤细胞中的生物学作用。方法在本研究中我们通过免疫组化检测了70对胶质瘤及正常脑组织中SLC1A5的表达;并分析其表达的临床病理学意义及预后意义。在体外胶质瘤细胞系U87中运用短发夹RNA干扰沉默SLC1A5后,检测U87细胞系的增生和浸润。结果相比正常脑组织,SLC1A5在胶质瘤中显著性上调表达;其表达水平与肿瘤大小及不良预后具有统计学相关性。沉默SLC1A5能够显著性抑制U87细胞的增生及浸润。结论 SLC1A5可以作为胶质瘤的一个潜在干预靶点。Objective Aerobic glycolysis has been taken as one of the most important metabolic ways of cancer cells.Recent studies showed that glutamine can also be used as key nutrient substance cancer cells depend on.Cancer cells could reprogram its metabolism driven by inside and outside factors,taking up extracellular glutamine.SLC1A5 was the most important glutamine transporter protein that can transport extracellular glutamine.Cancer cells thus could regulate the influx of glutamine through SLC1A5 to provide the necessary energy substance cancer cells need.SLC1A5 has been extensively reported in some malignant tumors.However,it remains largely unknown in glioma.The study aims to understand the clinicopathological significance of SLC1A5 expression and to explore its biological roles in glioma.Me thods 70 pairs of SLC1A5 of glioma and normal brain cells were tested by immunnohistochemistry to analyze their clinicopathological and prognostic significance.MTT and Transwell assays were used to assess the proliferative and migratory variation of U87 after SLC1A5 was successfully silenced using sh RNA.Re s ults Compared with normal brain cells,SLC1A5 was significantly up-regulated in glioma tissues and up-regulation of SLC1A5 was shown to be significantly correlated with poor prognosis and tumor size.Silencing of SLC1A5 was shown to be able to significantly suppress the proliferation and invasion of U87.Conclus ion SLC1A5 could be used as a promising target in the treatment of glioma.
