摘　　要：为探讨组蛋白去乙酰化酶抑制剂曲古抑菌素A（Trichostatin A,TSA）对刀豆素A（Concanavalin A,Con A）诱导的小鼠急性肝炎的保护作用及可能的作用机制,本研究利用Caspase 3活性试剂盒、流式细胞分析仪和酶联免疫法（ELISA）来分别检测Con A注射后12 h的小鼠肝组织中Caspase 3活性、CD4＋/CD69＋淋巴细胞比例变化和血清中干扰素（IFN）-γ、肿瘤坏死因子（TNF）-α水平。结果与模型组相比,TSA给药组小鼠血清转氨酶活性显著降低,IFN-γ、TNF-α水平明显降低;肝脏炎症损伤程度明显减轻,肝组织中Caspase 3活性显著降低。与模型组相比,TSA给药组小鼠肝组织中CD4＋/CD69＋比例显著降低。该研究表明曲古抑菌素A对刀豆素A诱导的急性肝炎具有保护作用,该作用可能与抑制肝脏T细胞介导的免疫反应有关。To investigate the protective effect and possible mechanism of histone acetylhydrolase inhibitor Trichostatin A（TSA） on Concanavalin A（Con A）-induced acute hepatitis in mice. The Caspase 3 activity kit,flow cytometric analyzer, and enzyme-linked immunosorbent assay（ELISA） were used to detect Caspase 3 activity,CD4＋/CD69＋lymphocyte ratio, and serum levels in liver tissue of mice 12 h after Con A injection. Interferon（IFN）-γ, tumor necrosis factor（TNF）-α levels. Results Compared with the model group, serum transaminase activity was significantly decreased in the TSA-administered group, and the levels of IFN-γ and TNF-α were significantly decreased. The degree of inflammation in the liver was significantly reduced, and the activity of caspase 3 in the liver tissue was significantly reduced. Compared with the model group, the proportion of CD4＋/CD69＋in the liver tissue of the TSA-administered group was significantly reduced. This study shows that trichostatin A has a protective effect against Concanavalin A-induced acute hepatitis, which may be related to inhibition of liver T cell-mediated immune responses.
