摘 要:鱼肉消化产物可在肠道内介导三价铁水解形成鱼水解物-铁氧化物纳米复合微粒(ANPs),但ANPs在肠道上皮细胞中的内吞作用机制及其对细胞铁代谢平衡的影响,目前尚不清楚。采用人结直肠腺癌细胞Caco-2构建了体外肠道上皮细胞模型,通过钙黄绿素荧光变化测定了细胞铁吸收,利用亚铁络合剂红菲绕啉二磺酸钠阻断了亚铁离子吸收途径。实验结果发现,阿米洛利和低温(4℃)对ANPs的内吞吸收有显著的抑制作用,叠氮钠和细胞外液缺钾则无显著影响;ANPs的内吞吸收呈现剂量依赖性和可饱和性,与硫酸亚铁相近。这表明肠道上皮细胞对ANPs的内吞吸收是一种依赖于细胞膜流动性、不依赖于ATP的吞饮过程,该过程受细胞铁稳态机制的调节,高浓度ANPs不会破坏肠道细胞铁代谢平衡。The intestinal digest of anchovy (Engraulis japonicus) has been found to mediate the luminal formation of ferric oxyhydroxide nanoparticles. However, the endocytic mechanism of anchovy hydrolysate-templated ferric oxyhydroxide nanoparticles (ANPs) in intestinal epithelial cells is still not clear yet. Calcein-fluorescence in polarized Caco-2 cell monolayers was used to monitor cellular iron uptake from ANPs, with the ferrous chelator bathophenanthroline disulfonate being employed to block the divalent metal transporter-l-mediated iron absorption. The results showed that iron absorption from ANPs was significantly inhibited by amiloride and low temperature (4 ℃), while sodium azide and potassium- deprivation exerted no significant effect. The iron absorption from ANPs was found to be dose-dependent and saturable, which was similar to than from ferrous sulfate. It thus seems that the endocytic uptake of ANPs was a process of pinocytosis depending on membrane fluidity, rather than adenosine triphosphate. Cellular iron uptake from ANPs was under the regulation of iron homeostasis, with hiqher doses of ANPs not disrupting cellular iron homeostasis.
