长链非编码RNA lnc-CDH11-3在胃癌组织中的表达及其干预效应分析

摘　　要：[目的]分析胃癌标本中长链非编码RNA lnc-CDH11-3的表达与临床病理特征的相关性,并在细胞水平上分析其对胃癌细胞的细胞活力、侵袭力及迁移能力的影响,探讨其在胃癌发生、发展中可能的调控及意义。[方法]收集46例胃癌组织及其癌旁正常组织标本,采用荧光实时定量PCR检测胃癌标本及癌旁正常胃黏膜组织中lnc-CDH11-3的表达量,并进一步分析临床病理特征与其表达水平的关系;利用慢病毒转染技术,建立lnc-CDH11-3表达量不同的胃癌稳定转染细胞株,运用CCK-8、Transwell跨膜实验检测细胞增殖活性、侵袭能力相关生物学功能;利用核浆分离及RT-PCR方法探究lnc-CDH11-3在细胞中的表达定位。[结果]Lnc-CDH11-3在癌组织的表达量高于所对应的癌旁正常胃黏膜组织（P〈0.05）。Lnc-CDH11-3表达水平与肿瘤分化程度、区域淋巴结转移、TNM分期相关;而与年龄、性别、肿瘤大小、部位等其他因素无明显联系。稳定转染胃癌细胞株,成功构建lnc-CDH11-3低表达细胞株,CCK-8增殖实验结果显示下调lnc-CDH11-3可显著降低胃癌细胞株的增殖能力;Transwell跨膜实验显示沉默lnc-CDH11-3可显著降低胃癌细胞株的侵袭能力。用RT-PCR技术检测发现lnc-CDH11-3主要表达于细胞核中,仅少许表达于胞浆中。[结论]Lnc-CDH11-3可作为促癌基因影响胃癌细胞增殖、侵袭等生物学特性,其表达量的变化与胃癌的发生发展密切相关。[Objective] To investigate the expression levels of lnc-CDH11-3 in gastric cancer（GC）and its relation to clinicopathological features of the tumor. [Methods] Forty-six specimens（cancer tissues and adjacent normal tissues） of GC were collected,the expression levels of lnc-CDH11-3 were detected by real-time RT-PCR,and its relationship with clinicopathological factors was analyzed. The gastric cancer BGC cells were transfected with lnc-CDH11-3 si RNA to silence the lncCDH11-3 expression. Cell proliferation and invasion were examined by CCK-8 assay and Transwell assay,respectively. Nucleocytoplasmic separation technology and RT-PCR were used to determine the intracellular localization of Inc-CDH11-3. [Results] The expression of lnc-CDH11-3 in GC tissue was upregulated in comparison with adjacent normal tissue（P〈0.05）. The expression of lnc-CDH11-3 was correlated with tumor differentiation,lymph nodes metastasis and TNM staging,but not correlated with age,gender,tumor size and tumor site. The CCK-8 assay showed that down-regulation of lnc-CDH11-3 significantly reduced the proliferation of BGC cells;and Transwell assay showed that silencing of Inc-CDH11-3 significantly reduced the invasive ability of BGC cells. RT-PCR found that lnc-CDH11-3 was mainly expressed in the nucleus,with only a little in the cytoplasm of GC cells. [Conclusion] lnc-CDH11-3 can promote proliferation and invasion of gastric cancer cells,and its expression levels are closely related to the clinicopathological features of gastric cancer.
