摘 要:目的探讨HOTAIR mRNA和EZH2 mRNA在肾细胞癌组织中的表达情况。方法选取行手术切除的经病理学检查确诊的肾细胞癌患者的肾细胞癌组织标本61例,同时选取对应的癌旁正常肾组织标本61例为对照;采用逆转录-聚合酶链式反应(RT-PCR)检测肾细胞癌组织和癌旁正常肾组织中HOTAIR mRNA和EZH2mRNA的表达水平。结果肾细胞癌组织中HOTAIR mRNA和EZH2 mRNA的相对表达量均明显高于癌旁正常肾组织(P﹤0.01);不同临床分期、淋巴结转移情况肾细胞癌患者的肾细胞癌组织中HOTAIR mRNA和EZH2mRNA的相对表达量比较,差异均有统计学意义(P﹤0.01);肾细胞癌组织中HOTAIR mRNA的相对表达量为(0.76±0.20),EZH2 mRNA的相对表达量为(2.04±0.51),两者呈正相关(r=0.683,P﹤0.01)。结论 HOTAIR mRNA和EZH2 mRNA在肾细胞癌组织中的表达水平明显高于癌旁正常肾组织,检测肾细胞癌患者的HOTAIR、EZH2水平对其肿瘤的恶性程度判断具有一定参考价值。Objective To investigate the expression ofHOTAIR mRNA and EZH2 mRNA in renal cell carcinoma tis- sue. Method Sixty-one renal cell carcinoma tissue specimens from patients with renal cell carcinoma confirmed by path- ological examination after surgical resection were selected, and 61 specimens of adjacent normal renal tissues were col- lected as controls. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression levels of HOTAIR mRNA and EZH2 mRNA in renal cell carcinoma tissues and adjacent normal renal tissues. Result The relative expression levels of HOTAIR mRNA and EZH2 mRNA in renal cell carcinoma tissues were significantly higher than those in adjacent normal renal tissues (P〈0.01). In renal cell carcinoma tissues of patients with different clinical stages and different lymph node metastasis, the relative expression levels ofHOTAIR mRNA and EZH2 mRNA were significantly different (P〈0.01); in renal cell carcinoma tissues, the relative expression ofHOTAIR mRNA was (0.76±0.20) and the relative expression of EZH2 mRNA was (2.04±0.51), which were positively correlated (r-=0.683, P〈0.01). Conclusion The expression levels of HOTAIR mRNA and EZH2 mRNA in renal cell carcinoma tissues were significantly higher than those in adjacent normal renal tissues. The detection of HOTAIR and EZH2 levels in patients with renal cell carcinoma has certain reference value for judging the malignant degree of tumors.
