摘  要:目的 探讨蛋白酪氨酸磷酸酶SHP-1高表达对食管鳞状细胞癌肿瘤细胞侵袭性和化疗敏感性的影响.方法 食管鳞状细胞癌EC9706细胞共分为3组,空白组未经任何处理;实验组采用SHP-1 mimic瞬时载体转染细胞24h,使用10 μmol/L顺铂再处理细胞12h;对照组采用10 μmol/L顺铂处理细胞12h.采用四甲基偶氮唑盐(MTT)法检测细胞增殖情况,Transwell小室法检测细胞侵袭性,实时荧光定量聚合酶链反应检测SHP-1 mRNA相对表达量,蛋白质印迹法检测SHP-1蛋白的表达水平.结果 实验组SHP-1 mRNA相对表达量(3.42±0.14)高于对照组与空白组(1.09±0.13、0.42±0.24,F=9.143,P< 0.05).实验组与对照组、空白组相比,细胞生长抑制率提高、细胞凋亡指数增加(均P<0.05),组间两两相比差异均有统计学意义(均P<0.05).实验组、对照组与空白组的细胞侵袭率分别为(6.5±1.3)%、(18.5±2.5)%和(45.2±7.2)%,差异有统计学意义(F=11.853,P<0.05).结论 SHP-1高表达能抑制食管鳞状细胞癌肿瘤细胞的侵袭性,提高化疗敏感性,促进细胞凋亡,抑制细胞增殖,可为提高食管鳞状细胞癌化疗疗效奠定一定的理论基础.Objective To investigate the effect of high expression of protein tyrosine phosphatase SHP-1 on the tumor cell invasiveness and chemosensitivity of esophageal squamous cell carcinoma.Methods EC9706 ceils of esophageal squamous cell carcinoma were divided into three groups:the blank group was not given any treatment;the experimental group used SHP-1 mimic instantaneous vector to transfect cells for 24 h,and used 10 μmol/L cisplatin to treat EC9706 cells for 12 h;the control group selected 10 μmol/L cisplatin to treat EC9706 cells for 12 h.Cell proliferation was detected by using methyl thiazolyl tetrazolium (MTT) assay,and cell invasion was detected by using Transwell chamber.The expression of SHP-1 mRNA was detected by using fluorescence quantitative polymerase chain reaction.The expression level of SHP-1 protein was detected by using Western blot.Results The expression of SHP-1 mRNA in the experimental group (3.42t0.14) was higher than that in the control group and the blank group (1.09±0.13,0.42±0.24,F =9.143,P 〈 0.05).Compared with the control group and the blank group,the cell growth inhibition rate and the apoptosis index in the experimental group were increased (both P 〈 0.05),and the differences between any two groups were also statistically significant (all P 〈 0.05).The cell invasion rate in the experimental group,the control group and the blank group was (6.5±1.3) %,(18.5±2.5) % and (45.2±7.2) %,respectively,and the difference was statistically significant (F =11.853,P 〈 0.05).Conclusion High expression of SHP-1 can inhibit the invasion of esophageal squamous cell carcinoma,improve chemosensitivity,promote apoptosis and inhibit cell proliferation,which could lay a theoretical foundation for improving the efficacy of chemotherapy for esophageal squamous cell carcinoma.
