针刺对颈椎间盘退变大鼠PGE_2、6-K-PGF1α和TXB_2含量的影响

摘　　要：目的:观察针刺对颈椎间盘退变大鼠的前列腺素E_2(prostaglandin E_2,PGE_2)、6-酮-前列腺素F(6-ketone Prostaglandin F,6-K-PGF1α)和血栓素B_2(throm-boxanep B_2,TXB_2)含量的影响。方法:对40只大鼠进行造模,造模成功后分为治疗组和模型组各20只,模型组造模后不做针刺处理。治疗组采用电针治疗,选穴:天柱穴(双),大椎,大柱穴(双),每次电针时间25 min,每日1次,1周为1个疗程,连续治疗3个疗程。另外取20只正常大鼠作为正常对照组。治疗结束后第2天,3组大鼠均采用戊巴比妥钠25 mg·kg-1腹腔注射麻醉处死,剪开颈部皮肤及肌肉,取出组织,检测PGE_2、6-K-PGF1α和TXB_2含量。结果:模型组PGE_2、6-K-PGF1α水平明显升高,与正常对照组比较有统计学意义(P<0.01),治疗组PGE_2、6-K-PGF1α、TXB_2含量与模型组比较有所降低,差异有统计学意义(P<0.05)。结论:针刺对大鼠椎间盘中PGE_2、6-K-PGF1α和TXB_2有明显的抑制作用。Objective： To observe the influence of acupuncture on contents of PGE2 and 6-K-PGF1α and TXB2 of rats with cervical intervertebral disc degeneration. Methods： After modeling,40 rats were divided into the treatment group and the model group,with 20 rats in each group. The ones in the model group did not take acupuncture treatment after having been modeled. The ones in the treatment group were treated with electroacupuncture. The chosen points were as follows： double Tianzhu（ BL 10）,Dazhui（ GV 14）,double Dazhu（ BI 11）. Each electroacupuncture time lasted for 25 minutes,once a day,one week as a course,and treatment lasted for 3 courses continuously. In addition,20 normal rats were taken into the normal control group. On the 2 nd day after the treatment,the 3 groups＇ rats were killed by intraperitoneal injection of excessive anaesthesia,the skin and muscles of the neck were cut off,the tissues were removed,and the contents of PGE2,6-k PGF1 and TXB2 were detected. Results： The levels of PGE2 and 6-k-PGF1α of the model group increased significantly There was a significant difference between the model group and the blank control group（ P〈 0. 01）. Compared with the model group,the contents of PGE2 and 6-k-PGF1αand TXB2 of the treatment group decreased,and the differences were statistically significant（ P 〈0. 05）. Conclusion： Acupuncture has an obvious inhibitory effect on inflammatory mediators including PGE2,6-k PGF1 and TXB2 in intervertebral disc.
