摘 要:目的采用正交试验设计优选测定奶制品中铬的石墨炉原子吸收光谱法的最佳仪器条件。方法利用正交试验考察灰化温度、灰化时间、原子化温度、原子化时间四因素对测定结果的影响。结果通过实验表明,原子化温度对测定影响最大,正交试验得到的最佳仪器条件为灰化温度1 400℃、灰化时间15 S、原子化温度2 250℃、原子化时间5 S。在优化的条件下得到的标准曲线方程为Y=0.0168 7X-0.004 7,r=0.999 3,方法的线性范围为0.00~20.0μg/L,检出限为0.054μg/L,加标回收率在93.5%~102.2%之间,相对偏差(RSD)小于5.0%。结论利用正交试验优选仪器分析条件,以较少的实验工作量获得较全面的信息,适用于石墨炉原子吸收法测定酸奶中铬的仪器条件的筛选。Objective The optimum conditions of the graphite furnace atomic absorption spectrometry with chromium in dairy products were optimized by orthogonal test. Methods The effects of four factors such as ash temperature, ashing time, atomization temperature and atomization time on the results were investigated by orthogonal test. Results The experimental results show that the atomization temperature has the greatest influence on the measurement, and the optimum conditions for the orthogonal test are the ashing temperature 1 400 ℃ Ashing time 15 S, atomization temperature 2 250 ℃, the atomization time 5 S.The standard curve equation obtained under optimal conditions is Y =0.01687 X-0.0047,r =0.999 3,The linear range of the method is0.00 -20.0 μg/L,Detection limit for 0.054 μg/L,The rate of recovery is between 93.5 % -102.2 %,The relative deviation(RSD) is less than 5.0 %. Conclusions The orthogonal test is used to optimize the instrumental analysis conditions and obtain more comprehensive information with less experimental workload. It is suitable for screening the instrument conditions for the determination of chromium in yogurt by graphite furnace atomic absorption spectrometry.
