摘  要:为研究急性抗阻运动后不同时相小鼠腓肠肌内Mighty基因表达和Notch信号通路的动态变化.将30只雄性5周龄C57BL/6小鼠,随机分为对照组(n=6)和运动组(n=24).对照组置于爬梯上自由活动,运动组分别于急性抗阻运动后即刻、6h、12h、24h取材.用RT-PCR检测肌合成抑制蛋白(myostatin,MSTN)和Mighty mRNA表达水平,用Western Blot检测NICD蛋白表达水平.RT-PCR结果显示,运动组小鼠运动后6hMighty出现显著性上调(p <0.05),运动后12hMighty达到峰值(p <0.05),运动后24hMighty有所下降,但仍具显著性差异(p <0.05);MSTN在运动后6h、12h出现显著性下调(p <0.05),运动后24h没有显著性变化(p>0.05).Western Blot结果显示,NICD在运动后即刻、6h出现显著性上调(p <0.05),运动后12h、24h没有显著性变化(p>0.05).急性抗阻运动可能通过激活Notch信号通路提高Mighty的基因表达水平.To investigate the dynamic change in the expression of Mighty gene and Notch signaling pathway at different time after acute resistance exercise. 30 C57BL/6 mice were divided into control group ( n=6) and exercise group ( n=24). Control group did free activity on crawling ladder. Exercise group were kiIIed at 0, 6, 12, 24 hours after acute resistance ex ercise. We tested the mRNA expression of myostatin and Mighty using RT PCR. Western Blot was used to study protein level of NICI). According to the date of the RT PCR, Mighty expression was increased significantly at 6 hours after exer cise ( p 〈0.05) , and reached its peak at 12 hours after exercise( p〈0.05) , then started to drop, however, which still had a significant difference (p〈0.05).Myostatin expression were down regulated significantly at 6, 12 hours after exercise ( p〈0.05) , however, there is no significant difference at 24 hours after exercise (p〉0.05). According Western Blot, NICI) protein level was increased significantly at immediately following exercise ( p〈0.05) and stayed elevated out to 6 hours ( p〈0.05), however, there is no significant difference at 12, 24 hours after exercise (p〉0.05). Acute resistance exercise could increase Mighty gene expression by activating Notch signaling pathway.
