摘  要:目的探究IL-8对乳腺癌MDA-MB-231细胞生物学特性的影响以及作用机制。方法将siRNA IL-8转染至乳腺癌细胞中,实验分为空白对照组、阴性对照组、siRNA1 IL-8组、siRNA2 IL-8组。噻唑蓝(MTT)实验观察干扰IL-8基因表达对细胞活力的影响,Transwell小室法检测细胞侵袭、迁移能力的变化,采用荧光定量PCR(RT-PCR)和酶联免疫吸附测定法(ELISA)检测转染后IL-8 mRNA和蛋白水平,蛋白质印迹法(Western blot)法检测Wnt/β-catenin信号通路的变化。结果与空白对照组相比,siRNA1 IL-8组、siRNA2 IL-8组乳腺癌细胞中IL-8 mRNA和蛋白表达量显著下降(P<0.05);siRNA2 IL-8组细胞中IL-8 mRNA和蛋白水平较siRNA1 IL-8组显著下调(P<0.05),表明siRNA2 IL-8的干扰效果较强,因此用于后续实验。干扰IL-8表达对抑制乳腺癌细胞的活力无显著影响,抑制其侵袭、迁移能力(P<0.05),细胞中β-catenin、MMP-2、MMP-9的蛋白水平显著降低(P<0.05)。结论干扰IL-8表达可能通过抑制Wnt/β-catenin信号通路从而抑制乳腺癌MDA-MB-231细胞侵袭、迁移能力。Interleukin-8(IL-8)was closely related to the occurrence and development of tumors,but its specific role in breast cancer cells is not fully understood. To investigate the effects of interleukin-8(IL-8)on the biological characteristics of breast cancer MDA-MB-231 cells and its mechanism. The siRNA IL-8 was transfected into breast cancer cells. The experiment were divided into blank control group,negative control group,siRNA1 IL-8 group,siRNA2 IL-8 group. The viability ofbreast cancer cells was measured by MTT assay. The invasion and migrationwere analyzed bytranswell assay. The levels of IL-8 were examined by real-time RT-PCR and ELISA. The activity of Wnt/β-catenin signal pathway were detected by Western blot. Compared with the blankcontrol group,the expression of IL-8 mRNA and protein in breast cancer cells in group siRNA1 IL-8 and siRNA2 IL-8 were decreased significantly(P〈0.05). The levels of IL-8 mRNA and protein in siRNA2 IL-8 group were significantly lower than those in siRNA1 IL-8 group(P〈0.05),indicating that siRNA2 IL-8 had strong interference effect,so it will be used for the subsequent experiments. The expression of interfering the expression of IL-8 had no significant effect on the viability of breast cancer cells,but inhibited invasion and migration ability of breast cancer cells(P〈0.05),reduced the protein levels of β-catenin,MMP-2,MMP-9(P〈0.05). Interfering the expression of IL-8 may inhibitthe invasion and migration of breast cancer MDAMB-231 cells by inhibiting the Wnt/β-catenin signaling pathway.
