摘 要:为了探索萃取-反萃分离纯化楸灵素的最佳条件,利用高效液相色谱(high performance liquid chromatography,HPLC)研究了溶剂种类、溶剂与物料的体积比以及萃取体系的pH值对分离纯化效果的影响。结果表明楸灵素在4种有机溶剂中油水分配系数大小依次是:乙酸乙酯>氯仿>二氯甲烷>甲苯;酸萃取时萃取剂体积过小,楸灵素存在降解;萃取-反萃过程中,随着pH增大,楸灵素在油水两相中分配比值减小。在pH=4.5发酵液-乙酸乙酯1∶1和pH=9氢氧化钠-乙酸乙酯1∶3条件下进行萃取反萃取,楸灵素的萃取收率达78.28%,对应楸灵素色谱纯度97.7%。本研究显示通过加入酸碱,调节电离平衡,改变其极性和油水两相分配系数分离纯化楸灵素的方法工艺是可行的。The influence of factors,such as the type of solvents,the ratio between solvent and broth(V/V)and pH,on the efficiency of separating and purifying catalitaxol,an effective anti-cancer drug,were studied in this paper.The partition coefficient of catalitaxol decreases when pH increases.Degradation of catalitaxol were observed when broth was extracted with too small volume of ethyl acetate at pH of 4.The yield of catalitaxol was 78.28%and the purity of catalitaxol was 97.7% when fermentation broth was extracted by ethyl acetate(volume ratio 1∶1)at pH of 4.5 and extracted reversely with water(volume ratio 3∶1)at pH of 9.These results suggest that it is feasible to separate and purify catalitaxol by adjusting its ionization equilibrium and polarity.
