含植物同源结构和环指域泛素样蛋白1在肝细胞癌中的表达及其临床意义

摘　　要：目的探讨含植物同源结构和环指域泛素样蛋白1（UHRFl）在肝细胞癌中的表达及其与肝癌患者预后的关系。方法免疫组织化学染色和荧光定量聚合酶链反应（PCR）技术检测UHRFl在50例肝细胞癌中的表达，分析UHRFl的表达水平与肝细胞癌患者预后的关系。结果33例肝细胞癌组织中UHRFlmRNA的表达水平较癌旁组织上调（P=0．001），32例肝细胞癌组织中UHRFl蛋白的阳性表达率高于癌旁组织（64％比22％，P=0．000），肝细胞癌组织中UHRFl的表达水平与肿瘤大小、TNM分期、转移及复发等病理参数相关，且UHRFl高表达的肝细胞癌患者的预后更差。结论肝细胞癌组织中UHRFl高表达提示预后不良。Objective To investigate the expression of ubiquitin -like with PHD and ring finger domains 1 （ UHRF1 ） in hepatocellular carcinoma （HCC） and its relationship with the prognosis of patients with HCC. Methods Immunohistoehemical staining and reverse transcription polymerase chain reaction （ RT - PCR） were used to detect the expression of UHRF1 in 50 HCC tissues, and the relationship between the level of UHRF1 expression and the prognosis of patients with HCC was analyzed. Results The expres- sion of UHRF1 mRNA in cancerous tissues was upregulated in 33 patients with HCC compared with para - cancerous tissues （P = 0. 001 ）, the positive expression rate of UHRF1 protein in cancerous tissues was al- so higher than that in para - cancerous tissues of 32 patients with HCC （64% vs. 22%, P =0. 000）, the expression level of UHRF1 were correlated with tumor size, TNM stage, tumor metastasis and relapse, and patients with HCC tended to have a worse prognosis. Conclusion Up - regulated expression of UHRF1 predicts poor prognosis of patients with HCC, and UHRF1 may be a good biomarker for evaluating the prog- nosis of patients with HCC.
