摘  要:目的观察抗瘤增效颗粒的急性毒性实验及对Lewis肺癌小鼠的抗肿瘤增效作用。方法将40只ICR小鼠(雌雄各半)随机等分为4组,分别为阴性对照组、抗瘤增效颗粒低剂量组、抗瘤增效颗粒中剂量组和抗瘤增效颗粒高剂量组,每组各10只(雌雄各半),分别予0.9%氯化钠注射液0.4 mL/10 g及等容积抗瘤增效颗粒溶液(生药含量分别为312、468、624 g/kg)灌胃,每日1次,连续2周。每次灌胃后4 h内密切观察各组小鼠外观、体征、行为活动,然后每日上午、下午观察外观、体征、行为活动及粪便性状等,并分别于灌胃前及灌胃7、14 d称各小鼠体质量;2周后解剖各组小鼠,肉眼大体观察各小鼠脏器及组织变化。另取C57BL-6小鼠制备Lewis肺癌模型,选取造模成功的小鼠40只,随机等分为模型组、顺铂(DDP)治疗组、抗瘤增效颗粒低剂量+DDP组、抗瘤增效颗粒中剂量+DDP组和抗瘤增效颗粒高剂量+DDP组,模型组予0.9%氯化钠注射液0.4 mL/10 g灌胃;DDP治疗组、抗瘤增效颗粒低剂量+DDP组、抗瘤增效颗粒中剂量+DDP组和抗瘤增效颗粒高剂量+DDP组均于第1、3、5 d予DDP溶液1 mL(含DDP 0.1 mg)腹腔注射,同时分别予0.9%氯化钠注射液、抗瘤增效颗粒溶液(生药含量分别为0.6、0.8、1 g/mL)0.4 mL/10 g灌胃,每日1次,连续21 d。观察比较5组抑瘤率、肿瘤体积及组织形态学变化。结果急性毒性实验结果显示,4组小鼠外观、体征、行为活动及粪便性状均未发现异常,灌胃前及灌胃7、14 d体质量未见明显异常变化,各脏器组织均未发现异常。药效学实验结果显示,与模型组相比,DDP治疗组、抗瘤增效颗粒低剂量+DDP组、抗瘤增效颗粒中剂量+DDP组和抗瘤增效颗粒高剂量+DDP组小鼠瘤质量均显著减轻(P<0.05),肿瘤体积均缩小(P<0.05),且抗瘤增效颗粒中剂量+DDP组和抗瘤增效颗粒高剂量+DDP组小鼠瘤质量低于DDP治疗组(P<0.05),抑瘤率高于DDP治疗组(P<0.05),肿�Objective To observe the Acute toxicity test of anti-tumor synergistic granule and its anti-tumor synergistic effect on Lewis lung cancer mice. Methods 40 ICR mice( male and female) were randomly divided into 4 groups,namely negative control group,anti-tumor synergistic granule low dose group,anti-tumor synergistic granule medium dose group and anti-tumor synergistic granule high dose group. Each group consisted of 10( male and female),respectively. The mice were given 0. 9% sodium chloride injection 0. 4 mL/10 g and the same volume of anti-tumor synergistic granule solution( the crude drug content was 312,468,624 g/kg respectively)once a day for 2 weeks,and the appearance,physical signs and behavioral activities of the mice in each group were closely observed within 4 hours after each administration.The appearance,signs and behaviors of the mice in each group were closely observed within 4 hours after the second administration,and then the appearance,signs,behavioral activities and fecal traits were observed every morning and af-ternoon. The weight of mice was measured before administration and 7 and 14 days after administration,and the changes of organs and tissues were observed with naked eyes after 2 weeks. Another 40 C57 BL-6 mice were randomly divided into model group,DDP treatment group,anti-tumor synergistic granule low dose + DDP group,anti-tumor synergistic granule medium dose + DDP group and anti-tumor synergistic granule high dose + DDP group.The model group was given 0. 9% sodium chloride injection 0. 4 mL/10 g daily. The DDP treatment group,anti-tumor synergistic granule low dose + DDP group,midium dose + DDP group and high dose + DDP group were all intraperitoneally injected by DDP solution 1 mL( containing DDP 0. 1 mg) on the first,third and fifth days. At the same time,0. 9% sodium chloride injection and anti-tumor synergistic granule( containing 0. 6,0. 8,1 g/m) were was administered 0. 4 mL/10 g every day for 21 d.
