摘  要:目的:测定淡豆豉中γ-氨基丁酸(GABA)及其前体物质谷氨酸(Glu)的含量。方法:通过单因素设计考察不同的提取方法、提取溶剂、提取时间、提取次数及料液比对淡豆豉中GABA、Glu提取率的影响,应用柱前在线衍生-高效液相色谱法测定其含量。结果:淡豆豉中GABA、Glu含量分别为5.48、17.98mg/g,最佳提取工艺条件为超声波水提法、料液比1∶12提取40min、提取2次,在0.0125~0.4000mg/mL范围内线性关系良好,平均加样回收率为分别为99.69%、97.34%,RSD(n=6)分别为4.99%、4.51%。结论:本研究首次发现淡豆豉中存在较高含量的GABA、Glu,高于目前已报道的大多数中药材和传统食用发酵豆制品,并建立了适合淡豆豉中GABA、Glu的提取工艺和柱前在线衍生-高效液相色谱检测法。Objective: To measure the contents of β-aminobutyric acid(GABA) and glutamic acid(Glu) in fermented soybean. Methods: It was investigated at the extraction methods, extraction time, extraction time, extraction times and ratio of solid to liquid on the extraction rate of GABA and Glu by single factor design. And the content was measured by pre-column online derivatization-high performance liquid chromatography(HPLC). Results: The content of GABA and Glu in fermented soybean were 5.48 mg/g and 17.98 mg/g. The optimum extraction conditions were ultrapure water extraction and the 1∶12 ratio of material to liquid for 40 min and twice, within the range of 0.0125-0.4000 mg/mL, the linear relationship was good and the average recoveries were 99.69%, 97.34%, and the RSD(n=6) were 4.99% and 4.51%, respectively. Conclusion: For the first time, the study finds that GABA and Glu in fermented soybean are higher than those in the traditional Chinese herbal medicines and traditional edible fermented soybean products, and the extraction technology of GABA and Glu in fermented soybean and the pre-column online derivatization-HPLC are established.
