摘  要:目的探讨沙利度胺对实验性自身免疫性脑脊髓炎(EAE)大鼠髓鞘、轴突的保护作用以及CD4^+ T细胞凋亡的影响。方法将30只雌性SD大鼠随机分为空白对照组、模型组和药物组,每组10只。利用豚鼠脊髓匀浆皮下注射方法制作EAE模型,自造模日起SD大鼠腹腔注射60mg/kg沙利度胺或无菌溶剂,共15d;至免疫诱导的第20天统一处死。坚牢蓝(LFB)染色、甘氨酸银染色分别观察髓鞘、轴突的损伤情况,免疫组化法检测星形胶质细胞、CD4^+ T细胞变化,流式细胞术检测外周血CD4^+ T细胞凋亡情况。结果与模型组比较,药物组髓鞘脱失、轴突丢失均有所缓解,星形胶质细胞增生反应明显减轻;而外周血凋亡CD4^+ T细胞比例明显升高,CD4^+ T细胞比例及中枢浸润CD4^+ T细胞水平均较低,差异均有统计学意义(P<0.05)。结论沙利度胺可减轻EAE大鼠髓鞘及轴突损伤,可能与促进CD4^+ T细胞凋亡有关。Objective To study the effect of thalidomide on the apoptosis of CD4^+ T cells in rats with experimental autoimmune encephalomyelitis(EAE). Methods Thirty female SD rats were randomly divided into blank group, control group and drug group with 10 rats in each group. EAE model was induced by subcutaneous injection of the guinea pig's spinal cord homogenate. Thalidomide( 60 mg/kg) or aseptic solvent was intraperitoneally injected in drug group and control group,respectively for 15 d, and the animals were sacrificed at d20. Myelin and axon damage was observed with LFB staining and glycine silver staining in spinal cord tissue. The changes of central astrocytes and CD4^+ T cells were detected by immunohistochemistry. Flow cytometry was used to detect the apoptosis of CD4^+ T cells. Results Compared with the control group, the demyelinating injury and the loss of axon were alleviated and the inflammatory reaction of astrocytes was also attenuated. The proportion of apoptotic CD4^+ T in drug group was higher than that of the control group(P〈0.05). Conclusion Thalidomide can reduce the injury of myelin and axon in EAE rats, which may be related to the promotion of CD4^+ T cell apoptosis.
