摘  要:目的基于网络药理学和生物信息学方法分析复方苦参注射液在治疗肝癌中的分子生物学机制。方法通过中药系统药理学数据库和分析平台筛选和预测复方苦参注射液的有效入血活性成分及其已知调控的基因,检索挖掘肝细胞癌的基因组数据库(Onco DB.HCC)中与肝癌相关联的基因。结合生物信息学方法,明确复方苦参注射液在治疗肝癌中对特异性基因的调节作用,运用Clue Go进行信号通路富集分析,分析其在治疗肝癌中可能发挥作用的分子生物学机制。结果检索出复方苦参注射液相关的成分185个,入血活性成分58个,可能的作用靶点146个;与人肝癌发生、发展相关的已知基因612个;信号通路30条。结论复方苦参注射液治疗肝癌具有多成分协同、调节多个基因的特点;其可能作用机制是通过调节肿瘤组织微环境,促肿瘤细胞凋亡、抑制其转移,还通过免疫炎症调节杀灭肿瘤细胞。与目前治疗肝癌的作用机制相一致。Objective To explore the molecular biological mechanism of compound matrine injection in the treatment of liver cancer based on network pharmacology and bioinformatics.Methods Screening and prediction of effective blood-intaking components of compound Kushen injection and its known regulatory genes were conducted through the pharmacological database and analysis platform of Chinese medicine system.Genes associated with liver cancer were searched for in the genome database for hepatocellular carcinoma(Onco DB.HCC).Combined with bioinformatics methods,it was clear that compound Kushen injection can regulate the specific genes in the treatment of hepatocellular carcinoma.Clue Go was used for signal pathway enrichment analysis to analyze the molecular biological mechanisms that may play a role in the treatment of hepatocellular carcinoma.Results A total of 185 components related to compound Kushen injection were obtained,and 58 active ingredients were injected into the blood and 146 targets were identified.A total of612 known genes related to the occurrence and development of liver cancer and 30 signal pathways were found.Conclusion Compound Kushen injection play roles of multi-component cooperation and regulation of multiple genes in the treatment of liver cancer by regulating the micro-environment of the tumor tissue,promoting tumor cell apoptosis,inhibiting its metastasis,and regulating the killing of tumor cells through immune inflammation.It is consistent with the current mechanism of action in the treatment of liver cancer.
