摘 要:目的探讨多柔比星联合蛋氨酸脑啡肽(Met-ENK)对神经母细胞瘤SH-SY5Y细胞的生长抑制及Tau蛋白磷酸化的调节作用。方法阳性对照组SH-SY5Y细胞以1. 25 mg·L^(-1)的多柔比星处理,低、中、高剂量实验组SH-SY5Y细胞分别以1. 25 mg·L^(-1)的多柔比星+10-7、10-5、10-3mol·mL^(-1)Met-ENK处理,阴性对照组细胞以等量生理盐水处理。MTT法检测各组SH-SY5Y细胞存活率,流式细胞术和Hoeschest 33258细胞核染色检测各组SH-SY5Y细胞凋亡情况,Western Blot法检测各组SH-SY5Y细胞p-Tau蛋白表达情况。结果与阴性对照组比较,干预24、48、72 h后,阳性对照组和各实验组细胞存活率降低,且实验组低于阳性对照组,实验组细胞存活率随着Met-ENK浓度的增大逐渐降低(P <0. 05)。与阴性对照组比较,阳性对照组和各实验组细胞凋亡率、G0/G1期细胞比例显著升高,中、高剂量实验组高于阳性对照组(P <0. 05);阳性对照组和各实验组S期细胞比例显著降低,中、高剂量实验组低于阳性对照组(P <0. 05)。与阴性对照组比较,阳性对照组和各实验组细胞p-Tau蛋白相对表达量明显升高,中、高剂量实验组高于阳性对照组(P <0. 05)。结论多柔比星联合Met-ENK可抑制神经母细胞瘤SH-SY5Y细胞生长,并促进其凋亡,这可能与上调Tau蛋白磷酸化水平有关。Objective To explore the effect of doxorubiein combined with methionine enkephalin (Met-ENK) on the growth inhibition and Tau protein phosphorylation of neuroblastoma SH-SY5Y cells. Methods SH-SY5Y cells in the positive control group were treated with 1.25 mg·L-1 doxorubiein, SH-SY5Y cells in the low, nfiddle, and high dose experimental groups were treated with 1.25 mg·L-1 doxorubiein + 10 7, 10 5, 10 3 mol·mL-1 Met- ENK, SH-SY5Y cells in the control group were treated with the same amount of normal saline. MTT assay was used to detect the proliferation of SH-SY5Y cells in each group, flow eytometry and Hoeehst 33258 staining were used to detect the apoptosis of SH-SY5Y cells, WB was used to detect the p-Tau proteins expression of SH-SY5Y cells in each group. Results Compared with the control group, after the cells intervented for 24, 48, 72 h, the cell viabili- ty of the positive control group and the experimental groups were decreased, and the experimental groups were lower than the positive control group, the viability of the experimental groups were decreased gradually with the increase of Met-ENK concentration ( P 〈 0.05 ). Compared with the control group, the apoptotie rates and percentage of G0/G1 phase cells in the positive control group and the experimental groups were significantly increased, and the middle and high dose experimental groups were higher than the positive control group ( P 〈 0.05 ) , the percentage of S phase cells in the positive control group and the experimental groups were significantly decreased, the middle and high dose experimental groups were lower than the positive control group (P 〈 0.05). Compared with the control group, the relative expression of p-Tau protein in the positive control group and the experimental groups were increased, the middle and high dose experimental groups were higher than the positive control group (P 〈 0.05).
