摘  要:目的探讨尼美舒利对胆管癌细胞Th1、Th2细胞因子分泌的影响。方法应用酶联免疫吸附试验检测胆管癌细胞HCCC-9810中白介素-2(IL-2)、IL-4、IL-6和干扰素γ(IFN-γ)等Th1、Th2细胞因子的表达,流式细胞仪检测尼美舒利对Th1、Th2细胞因子分泌的影响。结果尼美舒利作用后,随其药物浓度升高,胆管癌细胞HCCC-9810中IL-2、IFN-γ水平升高,而IL-4、IL-6水平下降,Th1/Th2比值升高(P <0. 05)。结论胆管癌细胞呈Th2优势状态,尼美舒利可能通过逆转Th1/Th2偏移调节细胞免疫,从而发挥抗肿瘤作用。Objective To explore the effect of nimesulide on secreting Thl and Th2 cell cytokines of cholangiocarcinoma cell line in vitro. Methods In the cholangiocarcinoma HCCC-9810 cells, interleukin-2 (IL-2) , IL4, IL-6 and interferon-γ (IFN-γ) were detected by enzyme-linked immunosorbent adsorption. The changes of Thl and Th2 cytokines were analyzed by flow cytometry. Results After being cultured with different concentration of nimesulide, IL-2 and IFN-γ concentration were higher, IL4 and IL-6 concentration were lower, and Th1/Th2 ratio was higher in the cholangiocarcinoma HCCC-9810 cells ( P 〈 0.05). Conclusion Cholangiocarcinoma cells showed Th2 dominance, nimesulide can convert the imbalance of Th1/Th2 cytokines, and regulate cellular immunity to prevent tumor growth.
