摘 要:目的了解P27^(Kip1)蛋白的表达与非小细胞肺癌(NSCLC)的关系。方法应用免疫组织化学SABC法检测43例NSCLC组织和27例肺良性病变组织中P27^(Kip1)蛋白的表达情况。结果 P27^(Kip1)蛋白表达可见于NSCLC的细胞核或细胞质。细胞核表达阳性率为9. 3%,显著低于肺良性病变组织的44. 4%(P <0. 05);细胞质表达阳性率为48. 8%,显著高于肺良性病变的7. 4%(P <0. 05)。P27^(Kip1)蛋白细胞核表达与细胞质表达的相关性分析示,两者表达呈负相关(r=-0. 313,P <0. 05)。P27^(Kip1)蛋白细胞核和细胞质表达与患者的临床病理特征均未见明显关系(P> 0. 05)。P27^(Kip1)蛋白细胞核表达阳性和阴性NSCLC患者术后3 a生存率分别为45. 0%和25. 0%,差异无统计学意义(P> 0. 05); P27^(Kip1)蛋白细胞质表达阳性和阴性NSCLC患者术后3 a生存率分别为33. 7%和50. 3%,差异无统计意义(P> 0. 05)。结论 P27^(Kip1)蛋白在NSCLC组织中表达的异常定位(细胞核表达减少,细胞质表达增多)是NSCLC患者预后不良的潜在因素之一。Objective To analyze the relationship between P27Kip1 expression and non-small cell lung cancer (NSCLC). Methods Immunohistochenlical SABC method was taken to detect the expression of P27Kip1 in the 43 cases of NSCLC and 27 cases of lung benign diseases. Results 9.3% of the NSCLC tissues was positive for nuclear P27Kip1 , lower than that of benign lung tissues (44.4% , P 〈 0.05 ). However, cytoplasmic P27Kip1 staining was observed in 48.8% of the NSCLC tissues, predominantly higher than that of the benign lung tissues (7.4% , P 〈 0.05 ). The correlation analysis showed that nuclear P27Kip1 tended to correlate negatively with cytoplasmic P27Kip1( r = -0. 313 ,P 〈 0.05 ). There was no significant difference between the expression of P27Kip1 and clinical characteristics (P 〉0.05). The 3-year survival rate in the nuclear P27Kip1-positive tumors was 45.0% , and was 25.0% in the nuclear P27Kip1 -negative tumors ( P 〉 0.05 ). The 3-year survival rate in the cytoplasmic P27Kip1 -positive tumors was 33.7% , and was 50.3% in the cytoplasmic P27Kip1 -negative tumors (P 〉 0.05). Conclusion Misloealization of P27Kip1 from nucleus to cytoplasm may play an important role in the pathogenesis of NSCLC.
