摘  要:目的:检测皮肤瘢痕癌组织中MDM2蛋白和mRNA的表达并探讨其作用机制。方法:以皮肤瘢痕癌、病理性瘢痕为研究对象,正常皮肤作为对照。采用免疫组化和原位杂交法,分别检测MDM2蛋白和mRNA的表达,运用图像分析系统检测3组标本的表达指标(平均光密度和阳性面积)。结果:在皮肤瘢痕癌组织中MDM2蛋白和mRNA的表达呈阳性或强阳性,在皮肤病理性瘢痕组织中呈弱阳性或阳性,在正常皮肤组织中呈阴性或弱阳性。组间两两比较差异均有统计学意义(P<0.05)。结论:皮肤瘢痕癌的发生可能与MDM2蛋白和mRNA的高表达有关。检测MDM2蛋白和mRNA表达水平,有可能预测皮肤病理性瘢痕早期癌变。OBJECTIVE: To investigate the mechanism of MDM2 mRNA and protein expression in the development of skin scar carcinoma. METHODS: Skin scar carcinoma and pathological scar were analyzed in comparison to normal skin. Expression of MDM2 protein was detected using immunohistochemical method and MDM2 mRNA using in situ hybridization. Their differential expressions (optical density and positive area) among the three groups of tissues were detected using computerized image-analysis. All data were input into computer and analyzed statistically using SPSS (17.0) software. RESULTS: Expressions of MDM2 mRNA and protein in skin scar carcinomas were positive or strongly positive, weakly positive or positive in skin pathological scar and negative or weakly positive in normal skin tissues. Differences between any two groups were statistically significant (P〈0.05). CONCLUSION: High expression of MDM2 mRNA and protein was significantly associated with development of skin scar carcinoma. Therefore, their expression may be useful for diagnosis of skin scar carcinoma in the earlier stages.
