摘 要:目的研究高脂饮食诱导过程中大鼠精子形态与线粒体功能损伤的动态变化特点。方法高脂饮食诱导肥胖大鼠模型,根据大鼠体重增加量分为肥胖组(DIO)(n=36)、肥胖抵抗组(DR)(n=36)和基础组(CON)(n=36)。DIO组和DR组喂高脂饲料,CON组则喂基础饲料。从第8 w起,每2 w,每组随机选取6只大鼠处死,提取附睾样品,测定相关指标。结果肥胖组精子密度相对于其他两组明显降低(P<0.05),但精子畸形数相对于其他两组明显升高(P<0.05)。在第10、12、18 w,DIO组膜电位(MMP)水平显著低于DR组,在第8、12、18 w时,DIO组显著低于CON组(P<0.05),在第16和18 w,DR组MMP水平显著低于CON组,(P<0.05)。5种线粒体复合体活性改变中线粒体复合体Ⅲ表现的最为明显,线粒体复合体Ⅰ和Ⅴ也表现出了活性降低的趋势。结论在高脂饮食诱导下,肥胖大鼠的精子密度降低、畸形数增高,线粒体出现损伤表现。Objective To observe the characteristics of sperm morphology and mitochondrial dysfunction in obese rats fed a high fat diet. Methods According to the gain of body weight, rats were divided into obese group (DIO)(n=36), obesity resistance group (DR) (n=36) and control group (CON) (n=36) after a 2 weeks high fat diet feeding. Then the DIO and DR groups were fed the high fat diet, while the control group was given the basic from diet. From the 8-week, 6 rats from every group would be killed every two weeks. The testis tissue was collected and the related parameters were measured. Results The weight of DIO group was higher than those in DR and CON groups (P〈0.05). Sperm density in DIO group was lower (P〈0.05), and the number of teratospermia was higher (P〈0.05). The membrane potential(MMP) was lower in DIO group at week 10,12,18 than that in DR group (P〈0.05), and at week 8,12,18, the MMP was lower than that in CON group(P 〈 0.05). The mitochondrial complex III changed most in the activity, which was decreased in the DIO group. The activity of mitochondrial complexes I and V also showed the decreasing trend. Conclusion Obesity induced by high-fat diet could cause decreased sperm density, increased teratospermia, and damaged function of mitochondria.
