摘 要:目的研究大蒜素(Allitridi)对体外人子宫内膜癌Ishikawa细胞顺铂化疗敏感性的影响并其机制。方法体外培养并取对数生长期Ishikawa细胞,设空白对照组、大蒜素(25μg/ml)组、顺铂(40μg/ml)组和联合干预[大蒜素(25μg/ml)+顺铂(20μg/ml)]组。MTT比色法测定细胞增殖抑制率,流式细胞术检测细胞周期并观察细胞凋亡状况,RT-PCR法测定Bax mRNA、Bcl-2 mRNA表达,Western blotting法检测激活型Caspase-3蛋白表达。结果较顺铂组,联合干预组细胞增殖抑制率显著升高,处于细胞周期G0/G1期比例显著升高而S期、G2/M期比例显著降低,细胞凋亡率(AI)显著升高,Bax mRNA表达上调、Bcl-2 mRNA表达下调且Bax/Bcl-2比值显著升高,激活型Caspase-3蛋白表达上调;较空白对照组,大蒜素组处于细胞G0/G1期比例显著提高、G2/M期比例显著降低,Bcl-2 mRNA表达下调、Bax/Bcl-2比值升高,激活型Caspase-3蛋白表达显著上调;上述差异均具有统计学意义(P<0.05,P<0.01)。结论大蒜素具有提高人子宫内膜癌细胞顺铂化疗敏感性的作用,机制可能与阻滞细胞周期及调节凋亡相关基因、蛋白表达有关。Objective To explore the sensibilization effect and mechanism of allitridi on Ishikawa cells in endometrial cancer treated with cisplatin. Methods In the culture in vitro,the Ishikawa cells at the logarithmic phase were collected. The blank control group,allitridi(25 μg/ml) group,cisplatin(40 μg/ml) group and the combined intervention group[allitridi(25 μg/ml) + cisplatin(40 μg/ml) ]were set up. The MTT colorimetric method was adopted to determine the cell proliferation inhibition. FCM was used to determine the cell cycle and observed the apoptosis rate. RT-PCR was applied to determine the expressions of Bax mRNA and Bcl-2 mRNA. Western blotting method was used to determine the expression of active caspase-3. Results Compared with the cisplatin group,the cell proliferation inhibition rate was increased significantly in the combined intervention group. The proportion of cell was increased significantly at G0/G1 phase,and that was reduced significantly at S phase and G2/M phase. The apoptosis index was increased significantly,the expression of Bax mRNA was up-regulated,Bcl-2 mRNA was down-regulated and Bax/Bcl-2 ratio increased significantly,and the active caspase-3 protein was up-regulated. As compared with the blank control group,in the allitridi group,the cell proportion at G0/G1 phase was increased significantly,that at G2/M phase was reduced significantly,the expression of Bcl-2 mRNA was down-regulated and Bax/Bcl-2 ratio increased,and the active caspase-3 protein was up-regulated significantly. All of the differences mentioned above were significant statistically(P〈0.05 or P〈0.01). Conclusion Allitridi improves the chemotherapy sensitization of cisplatin in Ishikawa cells in endometrial cancer treated with cisplatin. It is possibly related to the inhibition of cell cycle and the regulation of the relevant genes of apoptosis and protein expressions.
