摘 要:目的:采用复合多因素造模法,给予小鼠肥甘饮食、酒伤、劳役等方法并加以湿热外环境以建立湿热证小鼠模型,探讨湿热证中炎症反应与AQP之间的关系。方法:将40只雄性BALB/c小鼠随机分为正常对照组、脂多糖(LPS)组、湿热组、脾虚组,观察小鼠日常活动以及体征,测量小鼠每周体质量变化,采用HE染色检测舌部病理形态学变化。用酶联免疫吸附试验(ELISA)检测血清中LPS和肿瘤坏死因子-α(TNF-α)的浓度;免疫组化法检测结肠组织中核转录因子κBp65(NF-κBp65)、Toll样受体4(TLR4)、水通道蛋白质(AQP)3和水通道蛋白质(AQP)4的阳性表达。结果:与正常组相比,LPS组、湿热组、脾虚组中LPS、TNF-α、NF-κBp65、TLR4、AQP3的表达升高(P〈0.001),AQP4的表达下降(P〈0.001);与LPS组相比,湿热组、脾虚组LPS、TNF-α的含量下降(P〈0.001),湿热组NF-κBp65、AQP4的表达升高(P〈0.05),脾虚组TLR4、AQP3、AQP4的表达升高。结论:湿热证小鼠LPS、TNF-α、NF-κBp65、TLR4的水平增高与水通道蛋白AQP3、AQP4关系密切,而LPS的在其中的机制有待进一步深入研究。Objective: To use the method of compound multi-factor to give mice fat and sweet diet,wine injury and other methods and damp-heat environment in order to establish the damp-heat syndrome mice model,we studied the expressions of inflammatory factors and aquaporin. Methods: Forty mice were randomly divided into four groups: the normal control group,the lipopolysaccharide group,damp-heat syndrome group,spleen deficiency group. Based on the model of successful exploration mechanism of substance in the damp-heat syndrome,we observed the symptoms and signs of mice and body weight changes were measured weekly. We detected the changes of tongue morphology. ELISA was used to detect concentrations of LPS and TNF-α in the serum and immunohistochemical staining to detect NF-κBp65,TLR4,AQP3 and AQP4 protein expressions. Results: Compared with the normal group,the expressions of LPS,TNF-α,NF-κBp65,TLR4 and AQP3 in the model group wre significantly increased( P 〈0. 001),and the expression of AQP4 was decreased( P〈 0. 001). Compared with LPS group,the contents of LPS and TNF-α decreased in damp-heat syndrome group and spleen deficiency group( P〈0. 001). The expressions of NF-κBp65 and AQP4 in damp-heat syndrome group were increased( P 〈0. 05) and the expressions of TLR4,AQP3 and AQP4 in spleen deficiency group were increased. Conclusion: The increased levels of LPS,TNF-α,NF-κBp65 and TLR4 in damp-heat syndrome are closely related to AQP3 and AQP4 and the detailed mechanism remains to need further in-depth study.
