沉默AQP-1对PDGF诱导的视网膜色素上皮细胞增殖迁移的影响研究

摘　　要：目的研究沉默水通道蛋白1(AQP-1)对血小板源生长因子(PDGF)诱导的视网膜色素上皮细胞增殖迁移的影响。方法用PDGF处理视网膜色素上皮细胞hRPE,Realtime PCR和Western blot方法检测AQP-1表达变化。在hRPE细胞中转染AQP-1 siRNA慢病毒载体,经PDGF处理以后,Realtime PCR和Western blot检测细胞中AQP-1表达水平。MTT法检测细胞增殖,细胞爬片实验检测细胞迁移,Western blot检测细胞中MMP-2、MMP-9蛋白水平。结果PDGF处理后的hRPE细胞中AQP-1 mRNA和蛋白水平均升高,转染AQP-1 siRNA慢病毒载体后的hRPE细胞中AQP-1 mRNA和蛋白水平均降低。PDGF处理后的hRPE细胞增殖能力升高,细胞迁移能力也升高,细胞中MMP-2、MMP-9蛋白水平升高。沉默AQP-1后的hRPE细胞经PDGF处理后,细胞增殖能力降低,细胞迁移能力也降低,细胞中MMP-2、MMP-9蛋白水平降低。结论沉默AQP-1能够抑制PDGF诱导的视网膜色素上皮细胞增殖和迁移。Objective To study how silencing AQP-1 might affect proliferation and migration of retinal pigment epithelial cells induced by PDGF. Methods Retinal pigment epithelial cells （hRPE） were treated by PDGF. AQP-1 expression was detected by Realtime PCR and Western blot. AQP-1 siRNA lentiviral vector was transfected into PDGF-treated hRPE cells and AQP-1 expression was measured again. MTT assay was used to detect cell proliferation. Cell migration test was used to detect cell migration. Western blot was used to detect MMP-2 and MMP-9 expression. Results AQP-1 mRNA and protein increased in PDGF-treated hRPE cells but decreased after AQP-1 silencing. PDGF treatment increased proliferation and migration of hRPE cells with increased MMP-2 and MMP-9. Reversely, silencing AQP-1 decreased proliferation and migration of the cells as well as MMP-2 and MMP-9 levels. Conclusion Silencing AQP-1 can inhibit the proliferation and migration of retinal pigment epithelial cells induced by PDGF.
