摘  要:目的分析口腔鳞状细胞癌(OSCC)组织特征性长链非编码RNA(lncRNA)表达谱及lncRNA结肠癌相关转录子2(CCAT2)在疾病发生发展中的作用。方法回顾性将十堰市太和医院收治的73例OSCC患者作为研究对象,取其癌组织与癌旁正常组织,通过高通量lncRNA芯片对其lncRNA表达情况进行测定,对OSCC组织特征性lncRNA表达谱进行分析,进而利用实时荧光定量聚合酶链反应(RT-q PCR)法对其癌组织与癌旁正常组织CCAT2表达水平进行测定,进而分析CCAT2表达与OSCC临床病理特征的相关性。结果共检出lncRNA差异表达1 697个,在芯片lncRNA总数中占5.55%(1 697/30 586);其中,呈现表达下降754个,表达升高943个;共有21个lncRNA差异表达倍数至少为10倍,其中表达下降的lncRNA9个,表达升高的lncRNA12个。RT-q PCR检测结果发现,在OSCC癌组织中,ENST00000458468与UCAl表达较癌旁正常口腔黏膜组织显著升高(P<0.01)。OSCC癌组织中CCAT2表达量较癌旁正常组织明显升高(P<0.01)。不同肿瘤分化程度与TNM分期的OSCC患者CCAT2表达量的比较,均存在显著差异(P<0.01);其中,低分化、Ⅲ~Ⅳ期的患者CCAT2表达量较中/高分化、Ⅰ~Ⅱ期者显著升高(P<0.01)。而CCAT2表达量与性别、年龄、肿瘤大小、肿瘤部位及颈淋巴结转移均无显著相关性(P>0.05)。结论在OSCC癌组织中,诸多lncRNA出现表达异常,其中CCAT2在OSCC肿瘤组织中的表达量增多,提示可能参与OSCC的病理过程。Objective To analyze the tissue characteristic long non-coding RNA (lncRNA) expression profiles of oral squamous cell carcinoma (OSCC) and the role of lncRNA colon cancer associated transcript 2 (CCAT2) in the development of disease. Methods 73 cases of OSCC in our hospital were taken as the research object, and the cancer tissues and normal tissues adjacent to the cancer were taken. The expression of lncRNA was measured by high throughput lncRNA chip. The characteristic lncRNA expression profiles of the OSCC tissue were analyzed, and the real-time fluorescent quantitative polymerase chain reaction (RT-qPCR) method was used for the cancer tissue. The expression level of CCAT2 in normal tissues adjacent to cancer was measured, and the correlation between CCAT2 expression and OSCC clinicopathological features was analyzed. Results A total of 1 697 different expressions of lncRNA were detected in this group, which accounted for 5.55% (1 697/30 586) in the total number of chip lncRNA, in which the expression decreased by 754 and the expression increased by 943. There were 21 lncRNA differential expressions of at least 10 times, of which 9 were expressed as lncRNA, and the expression increased lncRNA12. RT-qPCR detection showed that the expression of ENST00000458468 and UCAl in OSCC cancer tissues was significantly higher than those in adjacent normal oral mucosa tissues ( P 〈0.01). The expression of CCAT2 in OSCC cancer tissues was significantly higher than that in adjacent normal tissues ( P 〈0.01). There were significant differences in the expression of CCAT2 in OSCC patients with different tumor differentiation and TNM staging ( P 〈0.01), and the CCAT2 expression in the patients with low differentiation and stage III to IV was significantly higher than that of medium / high differentiation and I to II ( P 〈0.01). There was no significant correlation between CCAT2 expression and sex, age, tumor size, tumor location and cervical lymph node metastasis ( P 〉0.05). C
