摘  要:目的探讨大建中汤对慢性炎性内脏痛大鼠的镇痛作用和可能作用机制。方法 40只SD大鼠随机分为正常组、模型组、对乙酰氨基酚组和大建中汤组各10只。除正常组外,其余各组均采用5%246-三硝基苯磺酸灌肠制备慢性炎性内脏痛大鼠模型。从造模结束后第2天开始大建中汤组给予大建中汤0.22 g/(kg·d)灌胃,对乙酰氨基酚组给予对乙酰氨基酚溶液0.002 g/(kg·d)灌胃,正常组和模型组灌胃生理盐水7 ml/(kg·d),各组均灌胃14天。记录各组大鼠一般情况及体重,实验结束后在20、40、60、80 mm Hg 4个扩张压力下进行大鼠腹壁撤回反射(AWR)评分,同时检测血清肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)含量以及结肠黏膜中白细胞介素1受体相关激酶4(IRAK-4)mRNA表达,HE染色观察结肠黏膜病理学情况。结果与正常组比较,模型组大鼠体重下降,AWR评分在各个扩张压力下均显著升高(P<0.01),结肠黏膜的病理损伤较明显,血清IL-6、TNF-α含量及结肠黏膜IRAK-4 mRNA表达升高(P<0.01)。与模型组比较,对乙酰氨基酚组、大建中汤组大鼠体重升高,AWR评分在各个扩张压力下均降低(P<0.05),结肠黏膜病理形态改善,血清IL-6、TNF-α含量及结肠黏膜IRAK-4 mRNA表达下降(P<0.01)。结论大建中汤对慢性炎性内脏痛大鼠有明显的镇痛作用,其机制与降低大鼠血清IL-6、TNF-α含量和结肠黏膜IRAK-4mRNA表达有关。Objective To study the analgesia effects and possible mechanism of Dajianzhong Decoction( 大建中汤) on rats with chronic inflammatory visceral pain. Methods A total of 40 SD rats were randomly divided into normal group,model group,acetaminophen group and Dajianzhong Decoction group,with 10 rats in each group. Except the normal group,the other 3 groups were clystered by 5% trinitrobenzene sulfonic acid( TNBS) in preparing the model of chronic inflammatory visceral pain rat. Dajingzhong Decoction( 0. 22 g/kg·d) was given to the Dajingzhong Decoction group since the second day after the modeling. The acetaminophen solution( 0. 002 g/kg·d) was given to the acetaminophen group by gavage. The saline solution( 7 ml/kg·d) was given to the normal group and the model group by gavage. The intervention for all groups last for 14 days continuously. The general conditions and body weight of the rats were recorded. The abdominal withdrawal reflex( AWR) score was performed under the pressure of20 mm Hg,40 mm Hg,60 mm Hg,and 80 mm Hg after the experiment. The content of tumor necrosis factor α( TNF-α) and interleukin-6( IL-6) in serum,interleuk-1 receptor-associated kinase 4( IRAK-4) mRNA expression on colonic mucosa was detected at the same time. The pathologic features of colonic mucosa were observed by HE staining.Results Compared with the normal group,the body weight of rates in model group was decreased and the AWR score was significantly increased under each kind of pressures( P〈0. 01). The pathological damage of the colonic mucosa was more obvious,and the levels of serum IL-6,TNF-α and the expression of IRAK-4 mRNA were significantly increased( P〈0. 01). Compared with the model group,the body weight of rates in acetaminophen group and Dajianzhong Decoction group were increased and the AWR score were decreased under each kind of pressures( P〈0. 05).
