摘 要:目的:探讨新鲜和冻干富血小板纤维蛋白(PRF)混合物对骨髓间充质干细胞(BMSCs)增殖和成骨能力的影响。方法:取新西兰大耳兔耳缘动脉血制备新鲜和冻干PRF,染色并进行组织学观察;分别收集新鲜、冻干和PRF混合物孵育1、4、7、14、21、28 d的释放液,再将其配置并分为冻干PRF组、新鲜PRF组、PRF混合组(F∶L=1∶1)和空白组4组,分别与兔BMSCs混合并于体外培养,于7、14 d检测各组的四甲基偶氮唑盐(MTT)值、碱性磷酸酶(ALP)活性及钙结节形成情况。结果:组织学观察显示,新鲜PRF呈疏松网状,表面光滑,纤维束排列紧密;冻干PRF呈海绵团块状,表面呈多孔状。PRF各实验组对BMSCs的增殖、ALP的活性及钙结节的形成均有促进作用(P <0. 05);其中,PRF混合组的促进作用明显优于新鲜PRF组和冻干PRF组(P <0. 05)。结论:PRF混合组更利于促进BMSCs的增殖和成骨分化。AIM: To study the effects of fresh and lyophilized PRF composite scaffolds on the proliferation and osteogenesis of bone marrow mesenchymal stem cells (BMSCs). METHODS: Fresh PRF and lyophilized PRF were prepared from the arterial blood of the ear margin of New Zealand white rabbit and were stained by histological staining. Rabbit BMSCs were isolated and their morphology and growth characteristics were observed. The fresh, lyophilized and mixed PRFof both were used for the culture of BMSCs in vitro. The MTT assay value, ALP activity and calcium nodule formation were examined at 7 and 14 d after culture respectively. RESULTS: The fresh PRF fibrin showed loose reticulate structure, and the lyophilized PRF fibrin was cavernous mass. HE examination showed that the surface of fresh PRF was smooth and the fiber bundles were arranged closely, and the surface of the lyophilized PRF was porous. In the fresh and lyophilized PRF composite scaffolds mixed group, the proliferation, the activity of ALP and the formation of calcium nodules of BMSCs were significantly higher than those of the other two groups ( P 〈0.05 ). CONCLUSION: Fresh and lyophilized PRF mixed hybrid scaffolds are more conducive to the proliferation and osteogenic differentiation of BMSCs.
